摘  要:目的:研究人参皂苷Rg3对人乳腺癌细胞株MCF-7增殖及凋亡的作用机制。方法:将人乳腺癌细胞株MCF-7作为研究对象,用CCK-8、流式细胞仪等方法检测人参皂苷Rg3对人乳腺癌细胞株MCF-7增殖及凋亡的影响。结果:人参皂苷Rg3对抑制人乳腺癌细胞株MCF-7的增殖作用在一定范围内具有时间和浓度呈正相关的特点,随着时间的延长及药物浓度的增加,增殖抑制的作用增强,且呈剂量依赖性关系,24 h、48 h、72 h的IC50值分别为210.26μg/mL、145.78μg/mL、91.78μg/mL。药物作用24 h后,G2期细胞百分比明显增加,G1、S期细胞百分比明显减少。随着人参皂苷Rg3浓度的增加,明显增强诱导细胞的凋亡(P<0.05)。结论:人参皂苷Rg3可对MCF-7细胞产生抑制作用,使细胞阻滞于G1期,并诱导细胞凋亡。Objective:To study the mechanism of ginsenoside Rg3 on the proliferation and apoptosis of human breast cancer cell line MCF-7.Methods:The human breast cancer cell line MCF-7 was used as the research object.The mechanism of ginsenoside Rg3 on the proliferation and apoptosis of human breast cancer cell line MCF-7 was detected by CCK-8 and flow cytometry.Results:The effect of ginsenoside Rg3 on the proliferation of human breast cancer cell line MCF-7 has a positive correlation with time and concentration within a certain range.With the extension of time and the increase of drug concentration,the effect on proliferation inhibition is enhanced.In a dose-dependent relationship,the IC50 values at 24 h,48 h,and 72 h were 210.26μg/mL,145.78μg/mL,and 91.78μg/mL,respectively.After 24 hours of drug action,the percentage of cells in G2 phase was increased significantly,and the percentage of cells in G1,S phase was decreased significantly.With the increase of ginsenoside Rg3 concentration,the apoptosis of induced cells was significantly enhanced.(P<0.05)Conclusion:Ginsenoside Rg3 can inhibit MCF-7 cells,block cells in G1 phase,and induce apoptosis.
