摘  要:目的分析弓形虫感染孕鼠血清蛋白的变化,寻找诊断先天性弓形虫病的蛋白分子。方法16只BALB/c孕鼠随机分成两组。实验组于孕8d灌胃接种弓形虫RH株速殖子5×10^4个/0.4ml只,致胎鼠患病。在孕18d取孕鼠血清,对照组和实验组采用ELISA方法检测鼠血清弓形虫循环抗原,通过双向电泳技术分离差异蛋白点,并对差异蛋白进行生物信息学分析;运用Uniprot数据库鉴定蛋白质种类,筛选适合诊断先天性弓形虫病的特异性蛋白。结果实验组与对照组比较存在86个差异蛋白点,其中34个蛋白表达量升高,52个蛋白表达量下降。选择4个差异性显著的蛋白(升高和降低各2个)进行分析,表达量降低的为结珠蛋白HPT_MOUSE和纤溶酶原PLMN_MOUSE,表达升高的为血液结合素HEMO_MOUSE和未知蛋白Q3UKU5_MOUSE。结论成功筛选了先天性弓形虫病孕鼠血清中差异表达的蛋白质,这些蛋白质表达的改变可能与疾病的发生和发展有关。Objectives To analyze the changes in serum proteins in pregnant mice infected with Toxoplasma gondii and to find protein molecules with which to diagnose congenital toxoplasmosis.Methods Sixteen BALB/c pregnant mice were randomly divided into two groups.On day 8of pregnancy,the experimental group was vaccinated with 5×10^4/0.4mL of tachyzoites of the RH strain of T.gondii via gavage,thus infecting fetal rats.Serum was collected from pregnant mice on day 18of pregnancy.ELISA was used to detect circulating antigens of T.gondii in the control group and the experimental group.Two-dimensional electrophoresis was used to separate differential protein spots.The differential proteins were analyzed bioinformatically.The Uniprot database was used to identify protein types and screen specific proteins in order to diagnose congenital toxoplasmosis.Results The experimental group and control group produced 86differential protein spots.The level of expression of 34proteins increased and the level of expression of 52proteins decreased.Four proteins with significantly different levels of expression(2increased and 2decreased)were selected for a-nalysis.Expression of globin HPT_MOUSE and plasminogen PLMN_MOUSE decreased and expression of blood binding hormone HEMO_MOUSE and unknown protein Q3UKU5_MOUSE increased.Conclusion Pregnant mice with congenital toxoplasmosis were successfully screened for differentially expressed proteins.The changes in the expression of these proteins may be related to the development and progression of congenital toxoplasmosis.
