孕期暴露PAHs对子代雄性大鼠生殖系统发育的影响

摘  要:目的研究孕期暴露多环芳烃类化合物(Polycyclic Aromatic Hydrocarbons,PAHs)对子代雄性大鼠生殖系统发育的影响。方法将18只SD孕鼠按随体重随机分为3组,分别为对照组(玉米油)、苯并(a)芘(benzo(a)pyrene,BaP)低剂量组(5mg/kg)和高剂量组(10mg/kg),于妊娠第12~19天采用灌胃染毒方式,建立暴露模型,染毒容量为2mL/kg,每天一次,连续1周。待母鼠自然分娩,测量哺乳期(出生后2周)子代雄性大鼠的体重;检测子代雄性大鼠在儿童期(出生后4周)、青春期(出生后9周)、成年期(出生后12周)时的体重、睾丸重量、血清中睾酮(testosterone,T)水平、精子数量和活力;HE染色观察睾丸形态学。结果与对照组相比,高、低剂量暴露组子代雄性大鼠在哺乳期、儿童期和成年期时的体重明显降低(P<0.01),睾丸重量明显减轻(P<0.01),T水平呈不同程度的下降(P<0.05),光镜下睾丸间质细胞(Leydig Cell,LC)集簇呈不同程度的减少;高、低剂量暴露组子代雄性大鼠在青春期、成年期时精子的数量、活动度和活动率呈明显的降低(P<0.01)。结论孕期暴露PAHs可影响子代雄性大鼠生殖系统发育。Objective To study the effect of exposure to polycyclic aromatic hydrocarbons(PAHs)during pregnancy on the development of the reproductive system of male offspring rats.Methods Eighteen SD pregnant rats were randomly divided into 3 groups according to body weight,namely control group(corn oil),benzo(a)pyrene(BaP)low dose group(5mg/kg)and high dose group(10mg/kg),and exposure models were established by intragastric exposure on the 12th to 19th days of pregnancy.The exposure volume was 2mL/kg,once a day for 1 week.After the mother mouse was naturally delivered,the weight of the offspring male rats was measured during lactation(2 weeks after birth).The body weight,testicular weight,testosterone(T)level in serum,sperm count and vitality of the offspring male rats were detected in childhood(4 weeks after birth),adolescence(9 weeks after birth),and adulthood(12 weeks after birth),respectively.HE staining was used to observe testicular morphology.Results Compared with the control group,the weight of progeny male rats in the high-dose and low-dose exposure groups during lactation,childhood,and adulthood was significantly reduced(P<0.01),the testicular weight was also significantly reduced(P<0.01),T level was shown varying degrees of decline(P<0.05).Under light microscope,Leydig Cell(LC)clustering was shown varying degrees of decrease.The number,activity and activity rate were significantly reduced in the male rats of high-dose and low-dose exposure groups during puberty and adulthood(P<0.01).Conclusion Exposure of PAHs during pregnancy can affect the reproductive system development of offspring male rats.
