摘 要:采用全基因组重测序结合Sequenom MassARRAY~■SNP技术,研究基质金属蛋白酶2(Matrix Metallopeptidase 2,MMP-2)基因g.23253382C>T、g.23250947C>T两个位点在多羔绵羊品种(湖羊、策勒黑羊和小尾寒羊)和单羔绵羊品种(萨福克羊、滩羊、苏尼特羊和草原型藏羊)中的多态性及其与小尾寒羊产羔数的关联。结果表明:MMP-2基因g.23253382C>T和g.23250947C>T位点在单、多羔绵羊品种中均存在CC、CT、TT三种基因型,且2个位点的基因型频率和等位基因频率在单、多羔品种间差异均不显著(P>0.05)。g.23253382C>T位点在单、多羔绵羊品种中均表现为低度多态(PIC<0.25);g.23250947C>T位点在策勒黑羊和苏尼特羊中表现为低度多态,在其余5个品种中表现为中度多态(0.25<PIC<0.5)。g.23253382C>T、g.23250947C>T两个位点在7个绵羊品种中均处于哈代-温伯格平衡状态(P>0.05)。关联分析表明,MMP-2基因位点g.23253382C>T、g.23250947C>T的不同基因型与小尾寒羊第1、2、3胎次的产羔数均无显著关联(P>0.05)。可见,MMP-2基因g.23253382C>T、g.23250947C>T不适于小尾寒羊产羔性状的选育。The experiment aimed to elucidate the polymorphisms of MMP-2 gene(g.23253382C>T,g.23250947C>T)in polytocous(Hu sheep,Cele black sheep and Small Tail Han sheep)and monotocous(Suffolk sheep,Tan sheep,Sunite sheep and Prairie Tibetan sheep) sheep breeds,and by using the whole-genome resequencing combined with Sequenom Mass ARRAY ~■ SNP technology,the association between the loci and the little size in Small Tail Han sheep was analyzed.The results showed that three genotypes were found at g.23253382C>T(CC,CT,TT),g.23250947C>T(CC,CT,TT) in all sheep breeds.Both genotype frequency and allele frequency at the two loci had no significant difference in all sheep breeds(P >0.05).The g.23253382C>T locus was at low polymorphism(PIC<0. 25) in polytocous and monotocous sheep. The g. 23250947C>T locus was at low polymorphism in Sunite sheep and Cele black sheep,it was at moderate polymorphism in other breeds(0.25<PIC<0.5).Among the 7 breeds,g.23253382C>T and g.23250947C>T were in Hardy-Weinberg equilibrium(P >0.05).The association analysis showed that the genotypes of g.23253382C>T and g.23250947C>T from the MMP-2 gene had no correlation with the litter size of first,second or third parity Small Tail Han sheep(P>0.05).In summary,g.23253382C>T and g.23250947C>T loci of MMP-2 gene might not be suitable for the breeding of prolific Small Tail Han sheep.
