摘 要:目的观察大黄饮片与大黄免煎颗粒单药、复方对血瘀模型大鼠血液流变学、凝血功能的影响。方法将60只SD大鼠随机分为对照组、空白组、大黄饮片组、大黄颗粒组、大黄复方汤剂组、大黄复方颗粒组各10只,除对照组外均构建血瘀大鼠模型;对照组、空白组、大黄饮片组、大黄颗粒组、大黄复方汤剂组、大黄复方颗粒组分别给予生理盐水、生理盐水、大黄饮片水煎液、颗粒水溶液、大黄复方汤剂、大黄复方颗粒溶液20 mL/kg灌胃,60 min后颈动脉取血并检测血液流变学[全血黏度(200 s)、全血黏度(50 s)、血浆黏度、红细胞聚积指数]、凝血功能[凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、血小板计数(PLT)、纤维蛋白原(FIB)]指标。结果血液流变学指标空白组﹥大黄饮片组、大黄颗粒组﹥大黄复方汤剂组、大黄复方颗粒组﹥对照组,P均<0.05;凝血功能指标中,PT、APTT空白组﹤大黄饮片组、大黄颗粒组﹤大黄复方汤剂组、大黄复方颗粒组﹤对照组,PLT、FIB空白组﹥大黄饮片组、大黄颗粒组﹥大黄复方汤剂组、大黄复方颗粒组﹥对照组,P均<0.05;血液流变学、凝血功能指标大黄饮片组与大黄颗粒组比较、大黄复方汤剂组与大黄复方颗粒组比较,差异均无统计学意义。结论大黄饮片与大黄免煎颗粒单药、复方均能显著改善血瘀模型大鼠血液流变学、凝血功能,复方优于单药;大黄饮片与大黄颗粒、大黄复方汤剂与大黄复方颗粒作用相似,可以相互替代。Objective To observe the effects of Dahuang decoction pieces and single drug and compound formulation of Dahuang decocting-free granules on the blood rheology and coagulation function of blood stasis model rats.Methods Sixty SD rats were randomly divided into the control group,blank group,Dahuang decoction pieces group,Dahuang granules group,Dahuang compound decoction group,Dahuang compound granules group,with 10 rats in each group.Blood stasis rat model was constructed except for the control group.Rats in above groups were given physiological saline,physiological saline,water decoction of Dahuang decoction pieces,granular solution,Dahuang compound decoction,Dahuang compound granules solution 20 mL/kg by gavage.After 60 min,blood was taken from the carotid artery to test the hemorheological indicators[whole blood viscosity(200 s),whole blood viscosity(50 s),plasma viscosity,red blood cell accumulation index]and coagulation function indicators[prothrombin time(PT),activated partial thromboplastin time(APTT),platelet count(PLT),and fibrinogen(FIB)].Results Hemorheological indexes were as follows:blank group﹥Dahuang decoction pieces group,Dahuang granules group﹥Dahuang compound decoction group,Dahuang compound granules group﹥control group,with statistically significant differences(all P<0.05).In the coagulation function indicators,PT and APTT were in the following order:the blank group﹤Dahuang decoction group,Dahuang granules group﹤Dahuang compound decoction group,Dahuang compound granules group,control group;PLT,FIB:blank group﹥Dahuang decoction group,Dahuang granules group﹥Dahuang compound decoction group,Dahuang compound granules group﹥the control group,with statistically significant differences(all P<0.05).There was no significant difference in hemorheology and blood coagulation function between Dahuang decoction group and the Dahuang granules group,and between Dahuang compound decoction group and the Dahuang compound granules group.Conclusion Dahuang decoction pieces and single drug and compound
