摘 要:目的探讨外源性予以重组生长转化因子11(r-GDF11)对SD大鼠动脉血管钙化的影响。方法将SD大鼠分为钙化组、溶媒组、低浓度组、中浓度组、高浓度组5组,适应性饲养1周后,每日分别予以腹腔注射生理盐水、r-GDF11溶媒液及低、中、高浓度r-GDF11,持续7周后开始诱导钙化。诱导钙化7周后取各组大鼠血液,分离其主动脉血管。采用Von kossa·HE染色观察血管钙化病理情况;采用ELISA法测定其血清中骨形态发生蛋白4(BMP4)、GDF11水平;采用Western blot法、免疫组化法测定血管中BMP4、成骨特异性转录因子2(RUNX2)等钙化标志蛋白及GDF11表达及分布。结果①钙化组及溶媒组血清BMP4、GDF11水平较低、中、高浓度组明显升高,且低、中、高浓度组血清BMP4、GDF11水平依次减少;②低、中、高浓度组较钙化组及溶媒组钙盐沉积减少,弹力纤维更为完整,且r-GDF11浓度越高,钙盐沉积越少,弹力纤维越完整;③低、中、高浓度组的钙化标志蛋白RUNX2、BMP4较钙化组及溶媒组表达降低,r-GDF11浓度越高,钙化标志蛋白表达越低,免疫组化中分布越稀少;r-GDF11浓度越高,组织中GDF11表达越高,免疫组化中分布越密集。结论外源性予以r-GDF11可以减轻SD大鼠动脉血管钙化。Objective To explore the effect of exogenous recombined-growth differentiation factor 11(r-GDF11)on calcification of arteries in SD rats.Methods The SD rats were divided into five groups:calcification group,solvent control group,low-concentration group,medium-concentration group and high-concentration group.After 1 week of adaptive feeding,intraperitoneal injection of normal saline,r-GDF11 solvent and low,medium,high concentration r-GDF11 were given daily for 7 weeks,and then calcification was introduced.After 7 weeks of calcification induction,blood was collected from each group and the aortic vessels were separated.Von kossa·HE staining was used to observe the pathological status of vascular calcification.ELISA was used to determine the GDF11 and bone morphogenetic protein 4(BMP4)level in serum.Western blot and immunohistochemistry were used to determine the levels of calcified markers like BMP4 and runt-related transcription factor 2(RUNX2)and the distribution of GDF11 in blood vessels.Results①The serum BMP4 and GDF11 levels in the calcification group and the solvent control group were significantly higher than those in the low,medium and high concentration group,and the serum BMP4 and GDF11 levels in the low,medium and high concentration group successively decreased.②Compared with the calcification group and the solvent control group,the calcium salt deposition in the low,medium and high concentration groups decreased,and the elastic fiber was more complete.The higher the r-GDF11 concentration,the less the calcium salt deposition and the more complete the elastic fiber.③The expression of RUNX2 and BMP4 in the low,medium and high concentration groups was lower than that in the calcification group and the solvent control group.The higher the r-GDF11 concentration,the lower the expression of calcification marker protein and the rarer the distribution of calcification marker protein in the immunohistochemistry.The higher the concentration of r-GDF11,the higher the expression of GDF11 in tissues and the mor
