摘 要:为三七绿紫茎三萜皂苷合成转录调控机理提供参考,将彻底长成的绿紫茎均分为18段,用实时荧光定量PCR检测茎段的三七碱性螺旋-环-螺旋蛋白基因(bHLH)、乙烯响应因子基因(ERF)、v-myb骨髓母细胞增多症病毒癌基因同源物基因(MYB)和WRKY1蛋白基因(WRKY1)的转录水平,用香草醛-高氯酸比色法检测茎段的总三萜皂苷含量(TTSC),分析转录水平与TTSC间的Pearson相关性。结果表明:从三七绿紫茎的顶部到基部,三七bHLH、ERF、MYB和WRKY1的转录水平分别呈波动式平缓上升、不规则7峰、波动幅度逐渐增大的6峰和具2个主峰的6峰曲线;TTSC呈V型曲线,最低TTSC出现在茎上部的黄金分割点处;4个转录因子基因的转录水平和TTSC在不同茎段间的差异均达到极显著水平,且转录水平与TTSC之间的Pearson相关系数分别为0.371、0.130、-0.169和-0.195。因此,与三七绿紫茎三萜皂苷合成相关的主要转录因子应为三七的碱性螺旋-环-螺旋蛋白,其次为乙烯响应因子。In order to provide a reference for exploring the synthetic transcription regulation mechanism of the triterpenoid saponin in the purplish-green stems of Panaxnotoginseng,the fully grown purplishgreen stems were evenly divided into 18 segments,the transcriptional levels of the basic helix-loop-helix protein gene(bHLH),ethylene response factor gene(ERF),v-myb myeloblastosis viral oncogene homolog gene(MYB)and WRKY1 protein gene(WRKY1)of P.notoginsengin the segments were determined by using quantitative real-time PCR(qRT-PCR),the total triterpenoid saponin contents(TTSCs)of the segments were determined by using vanillin-perchloric acid colorimetric method and the Pearson correlations between the transcriptional levels and the TTSC were analyzed as well.Results:From the tops to the bases of the purplish-green stems of P.notoginseng,the transcriptional levels of the bHLH,ERF,MYB and WRKY1 displayed a"fluctuating and gently rising curve","irregular sevenpeaked curve","six-peaked curve with gradually increasing fluctuations"and"six-peaked curve with two main ones",respectively,whereas the TTSC showed a"V-shaped"curve,being characterized by the lowest TTSC at the golden section points in the upper parts of the stems.The differences of the transcriptional levels of the four transcription factor genes and TTSCs among different segments all reached extremely significant level,and the Pearson correlation coefficients between the transcriptional levels and the TTSC were 0.371,0.130,-0.169 and-0.195,respectively.Consequently,the main TFs related to the triterpenoid saponin synthesis in the purplish-green stems of P.notoginsengshould be the basic helix-loop-helix protein(bHLH),followed by the ethylene response factor(ERF).
