大鼠80%门静脉结扎残肝组织中CyclinD1和PCNA的表达与肝再生的关系分析

摘　　要：目的探究大鼠80%门静脉结扎残肝组织中周期蛋白D1(CyclinD1)和增殖细胞核蛋白抗原(PCNA)的表达与肝再生的关系。方法选取雄性SD大鼠144只(72只做生存率分析,72只做剩余实验),体重(220±20)g,随机分成3组PVL组、PH组、Sham组,每组24只。PVL组:行80%肝门静脉结扎;PH组:行80%肝切除;Sham组:仅翻动肝脏各分叶。比较各组大鼠术后生存率;分别于术后24h、48h、72h、120h每组各处死6只大鼠,检测其肝重量;检测各组血清ALT、AST含量水平;蛋白质印记法检测各组CyclinD1表达情况;免疫组化染色检测各组PCNA阳性率。结果生存率实验结果显示PVL组大鼠120h生存率为87.50%显著高于PH组62.50%,差异有统计学意义(P<0.05);术后24h,相比PVL组,PH组大鼠肝脏指数升高(P<0.05),48h、72h、120h肝脏指数均显著降低(P<0.05),相比PH组Sham组各时间段肝脏指数均显著降低(P<0.05);24h、48h、72h时,相比PVL组,PH组大鼠ALT和AST含量水平均显著升高(P<0.05),Sham组上述时间段均显著降低(P<0.05),相比PH组Sham组上述时间段ALT和AST含量水平均显著降低(P<0.05);120h时各组大鼠ALT和AST含量水平均无显著差异(P>0.05);术后24h,相比PVL组,PH组大鼠CyclinD1蛋白相对表达水平显著升高(P<0.05),48h、72h、120h时,相比PVL组,PH组大鼠CyclinD1蛋白相对表达水平显著降低(P<0.05);免疫组化结果显示,相比PH,PVL组大鼠PCNA蛋白阳性率显著升高(P<0.05)。结论大鼠80%肝脏门静脉结扎不仅可提高生存率同时还会促进相关增殖因子CyclinD1和PCNA的表达促进残余肝脏的再生。Objective To explore expression of cyclin D1(CyclinD1)and proliferating cell nuclear antigen(PCNA)in rat residual liver tissues of 80%portal vein ligation(PVL)and their relationship with liver regeneration.Methods A total of 144 male SD rats(72 cases for survival analysis,the other 72 cases for remaining experiments)weighing(220±20)g were randomly divided into PVL group,PH group and Sham group,24 cases in each group.PVL group underwent 80%hepatic PVL.PH group underwent 80%hepatectomy.In Sham group,only all liver lobulation was turned over.Postoperative survival rates of rats in each group were compared.At 24h,48h,72h and 120h after surgery,6 rats in each group were sacrificed.The liver weight was detected.The levels of serum ALT and AST in each group were detected.The expression of CyclinD1 in each group was detected by Western blotting.The positive rate of PCNA in each group was detected by immunohistochemical staining.Results Survival rate experiment results showed that the 120h survival rate in PVL group was significantly higher than that in PH group(87.50%vs 62.50%)(P<0.05).Compared with PVL group,at 24h after surgery,liver indexes in PH group were increased(P<0.05),while at 48h,72h and 120h after surgery,liver indexes were significantly decreased(P<0.05).Compared with PH group,liver indexes were significantly decreased in Sham group(P<0.05).At 24h,48h and 72h after surgery,compared with PVL group,levels of ALT and AST in PH group were significantly increased(P<0.05),while the above indexes in Sham group were significantly decreased(P<0.05).Compared with PH group,levels of ALT and AST in Sham group were significantly decreased(P<0.05).There was no significant difference in ALT or AST level among all groups at 120h after surgery(P>0.05).At 24h after surgery,compared with PVL group,relative expression level of CyclinD1 protein in PH group was significantly increased(P<0.05).At 48h,72h and 120h after surgery,compared with PVL group,relative expression level of CyclinD1 protein in PH group was significantly decr
