环磷酰胺诱导气虚体质大鼠模型的探索性研究

摘　　要：目的:以环磷酰胺腹腔注射联合饮食睡眠剥夺方法建立气虚型中医体质大鼠,并观察补中益气汤灌胃对模型的反证效果。方法:将40只大鼠随机分成空白组、饮食睡眠剥夺组、环磷酰胺组、中药组,每组10只。空白组每日正常饲养;饮食睡眠剥夺组每日放置在自制的游泳盒中进行饮食睡眠剥夺;环磷酰胺组在饮食睡眠剥夺组的基础上按照80mg/kg连续注射环磷酰胺3天,以后按照40mg/kg每周注射1次进行维持;中药组在环磷酰胺组的基础上每日灌胃中药(1ml/100g),实验周期为2周。观察各组大鼠的一般行为学表现,判断各造模方法与中医气虚体质的相似度。比较各组大鼠的血液学指标(RBC、HGB、HCT、PLT),免疫学指标(WBC、IgA、IgG、IgM),糖、脂肪、蛋白质代谢指标(GLU、Tch、TG、HDL、LDL、TP、ALB、GLO、A/G、ALT、AST),胸腺、脾脏指数及基础代谢率。结果:大鼠一般行为学观察发现,环磷酰胺组更接近气虚型中医体质的评定标准。与空白组比较,饮食睡眠剥夺组血液检查及脏器指数未出现统计学差异(P>0.05),环磷酰胺组血液检查(ALT、AST升高)及脏器指数均出现降低(P<0.05),与环磷酰胺组比较,中药组血液检查(ALT、AST降低)及脏器指数均出现升高(P<0.05)。与空白组比较,其他3组大鼠基础代谢率均出现不同程度降低,其中以环磷酰胺组最为明显,各组之间存在统计学差异(P<0.05)。结论:环磷酰胺腹腔注射联合饮食睡眠剥夺能够较好的复制气虚型中医体质模型,并可以通过补中益气汤反证。Objective:To establish traditional Chinese medicine constitution rats with Qi-deficiency by intraperitoneal injection of cyclophosphamide combined with dietary sleep deprivation,and to observe the counter evidence effect of Buzhong Yiqi Decoction on the model.Methods:The rats were randomly divided into blank group,sleep deprivation group,cyclophosphamide group and Chinese medicine group,10 rats in each group.The blank group was fed normally every day.The dietary sleep deprivation group was placed in a homemade swimming box for daily dietary sleep deprivation.In the cyclophosphamide group,80 mg/kg of cyclophosphamide was injected continuously for 3 days on the basis of the diet and sleep deprivation group,and then 40 mg/kg was injected once a week for maintenance.On the basis of cyclophosphamide group,traditional Chinese medicine group was given Chinese medicine by intragastric administration every day,1 ml/100 g,and the experimental period was 2 weeks.The general behavior of rats in each group was observed to determine the similarity between the modeling methods and the constitution of Qi-deficiency in traditional Chinese medicine.Blood parameters(RBC,HGB,HCT,PLT),immunological parameters(WBC,IgA,IgG,IgM),glucose,fat,protein metabolism parameters(GLU,Tch,TG,HDL,LDL,TP,ALB,GLO,A/G,ALT,AST),thymus,spleen index and basal metabolic rate were compared in each group.Results:The observation of general behavior in rats showed that the cyclophosphamide group was closer to the evaluation standard of TCM constitution of Qi-deficiency.Compared with the blank group,diet,sleep deprivation set of blood tests and viscera index was no statistical difference(P>0.05),blood test(ALT,AST)and viscera index of the cyclophosphamide group were lower(P<0.05),compared with the cyclophosphamide group,the blood tests(ALT,AST)and viscera index of the Chinese medicine group are higher(P<0.05).Compared with the blank group,the basal metabolic rate of the other three groups all decreased to different degrees,among which the cyclophosphamide grou
