辽藁本内生细菌ZHAB63鉴定与拮抗菌筛选

摘　　要：采用组织块法,从健康辽藁本中共分离出64株内生细菌,经初筛和复筛,菌株ZHAB63对11种常见中药材病原菌拮抗率均在50.00%以上(P<0.01),根据菌株形态特性、16SrDNA基因序列测定以及系统发育树初步鉴定发现,菌株ZHAB63为解淀粉芽孢杆菌(Bacillus amyloliquefaciens);采用L9(34)正交设计试验优化该菌株发酵培养基及发酵条件,确定液体发酵最适培养基为:蔗糖15.0 g·L^-1,酵母粉20.0 g·L^-1,蛋白胨20.0 g·L^-1,硫酸镁5.0 g·L^-1;最佳发酵条件:温度30℃,pH 6.5,转速180 r·min-1,培养时间48 h。研究表明,辽藁本中内生细菌资源丰富,以其研制新配方生物农药有待深入开发。Using the tissue block method,64 endophytic bacteria were isolated from L.jeholense.One strain ZHAB63,which the antagonism rates were above 50.00%on 11 pathogenic bacteria,was obtained through preliminary screening and rescreening.The strain ZHAB63 was identified as Bacillus amyloliquefaciens according to the morphological,16SrDNA gene sequence determination and phylogenetic tree.The L9(34)orthogonal design test was used to optimize the fermentation medium and fermentation conditions of the strain.The optimal medium for ZHAB63 growth was sucrose 15.0 g·L^-1,yeast powder 20.0 g·L^-1,peptone 20.0 g·L^-1,and magnesium sulfate 5.0 g·L^-1;and the optimal fermentation conditions were temperature 30℃,pH 6.5,rotation speed 180 r·min-1,and culture time 48 h.The research proved that the endogenous bacterial resources of L.jeholense were abundant,which could provide a theoretical basis for the development of new formula biological pesticides and was worthy of in-depth development.
