摘  要:为探究在奶牛乳腺发育分化中细胞-ECM相互作用及相关整联蛋白β1信号机制,构建包被不同蛋白成分(BSA、LN、Matrigel)作为细胞培养底物的transwell培养模型,奶牛乳腺上皮细胞在分化培养液诱导下,检测整联蛋白β1及其下游信号分子ILK与Rac1表达变化。通过添加AIIB2抗体调控整联蛋白β1活性,分析下游信号分子ILK与Rac1表达变化。结果表明,LN可上调整联蛋白β1-ILK-Rac1途径提高β-酪蛋白合成分泌,AIIB2阻断可下调整联蛋白β1下游信号分子ILK表达,抑制β-酪蛋白合成分泌。研究发现,在分化培养液诱导下,LN可上调奶牛乳腺上皮细胞β-酪蛋白合成分泌,这种上调作用依赖整联蛋白β1-ILK通路活性。In order to explore the interaction of cell-ECM and the mechanism of integrinβ1 in the development and differentiation of bovine mammary gland.In this study,a transwell culture model with different protein components(BSA,LN,and Matrigel)as cell culture substrate was constructed to detect the expression of integrinβ1 and its downstream signal molecules ILK and Rac1 in BMECs under the induction of differentiation medium.The expression of downstream signal molecules ILK and Rac1 was detected by adding AIIB2 antibody to regulate the activity of integrinβ1.The results showed that LN could up-regulate the integrinβ1-ILK-Rac1 pathway and enhance the synthesis and secretion ofβ-casein,AIIB2 block could down down-regulate the downstream signal molecule ILK expression of ILK downstream of integrinβ1 and inhibit the synthesis and secretion ofβ-casein.It was suggested that LN could up-regulate the synthesis and secretion ofβ-casein induced by differentiation medium,and this up regulation which depends depended on the activity of integrinβ1-ILK pathway.
