摘　　要：目的:探讨八肽胆囊收缩素受体(CCK-R)在八肽胆囊收缩素(CCK-8)改善脂多糖(LPS)诱导的内毒素血症(ETM)大鼠肺损伤中的作用。方法:SD大鼠随机分为4组(n=6):对照组(LPS+CCK-8组)、CCK-1受体(CCK-1R)阻滞剂组、CCK-2受体(CCK-2R)阻滞剂组、DMSO+PF组。尾静脉注射LPS(5 mg/kg)后,依次给予CCK受体阻滞剂(0.5 mg/kg,LPS后20 min)、CCK-8(20μg/kg,LPS后30 min)。记录潮气量(TV);观察肺组织病理学变化;检测肺组织匀浆中炎症因子(TNF-α、IL-1β和IL-6)的含量。结果:与对照组相比,CCK-2受体阻滞剂组大鼠潮气量减低,肺组织损伤程度明显加重,肺组织TNF-α、IL-1β和IL-6含量明显增加。结论:CCK-2受体在CCK-8治疗ETM大鼠肺损伤中起主要作用。Objective:To investigate the effects of CCK-8 receptor on lung injury in endotoxemia rats.Methods:Male SD rats were randomly divided into four groups(n=6)control group(LPS+CCK-8 group),CCK-1R antagonist group,CCK-2R antagonist group,DMSO+PF group.The rats were injected by LPS(5 mg/kg).CCK-8(20μg/kg)was administered 30 min after LPS injection.Either a specific CCK-1R antagonist or CCK-2R antagonist(0.5μg/kg)was injected before CCK-8 treatment(after LPS 20 min).The tidal volume was collected by a multi-channel data physiological recorder.The lung injury was observed by light and electron microscopy.The concentrations of TNF-α,IL-1βand IL-6 in lung homogenates were measured by ELISA kits.Results:Compared with control group,the tidal volume were significantly lower and the lung injuries were more serious in CCK-2R antagonist group.As well as the concentrations of TNF-α,IL-1β,and IL-6 in lung homogenates were higher.Conclusion:CCK-2 receptor plays a major role in the effect of CCK-8 on lung injury in ETM rats.
