龙胆泻肝汤对人HaCaT细胞STAT3信号通路的调控研究

摘　　要：目的确定龙胆泻肝汤对人角质形成细胞HaCaT细胞内STAT3通路的影响,探讨JAK-STAT3信号通路与龙胆泻肝汤治疗银屑病机制的相关性。方法使用Western blot检测细胞STAT3蛋白的表达情况,实时定量聚合酶链反应(PCR)检测STAT3 mRNA的表达,荧光素酶报告基因检测STAT3的转录功能。结果龙胆泻肝汤药物处理组HaCaT细胞STAT3总蛋白以及磷酸化蛋白水平均有明显下降,细胞核内的STAT3的表达也有明显降低;龙胆泻肝汤作用后HaCaT细胞内STAT3的转录功能明显下降;实时定量PCR结果显示龙胆泻肝汤处理细胞内STAT3的mRNA略有升高。结论龙胆泻肝汤能有效抑制HaCaT细胞内STAT3信号通路的活性,其机制可能与转录后水平抑制STAT3的表达有关。Objective In order to characterize the effects of Longdan Xiegan recipe on STAT3 passway in HaCaT cells.Methods Cells were treated with Longdan Xiegan recipe and protein expression was detected by Western blot,mRNA expression was measured by real-time polymerase chain reaction(PCR),and STAT3 transcription activity was measured by luciferase reporter assay.Results Longdan Xiegan recipe treatment significantly decreases the levels of total and phosphorylated STAT3 from whole cell lysate.In addition,Longdan Xiegan recipe downregulated expression of nuclear STAT3 and its transcription activity.However,results of real-time PCR showed that the mRNA levels of STAT3 was not decreased but slightly increased in Longdan Xiegan recipe treated cells.Conclusion Longdan Xiegan recipe could inhibit STAT3 passway in HaCaT cells,which may be related to downregulation of STAT3 expression at post-transcription levels.
