摘  要:目的建立增生性瘢痕的有效动物模型。方法于SD大鼠鼠尾形成6 mm矩形创面,根据创面边缘距鼠尾根部的距离分为A组(3 cm)和B组(5 cm)。术后16 d进行牵拉并测量创面牵张力。于牵拉7、14、21、28 d拍照记录两组的瘢痕的大体变化。于牵拉28 d检测两组瘢痕中张力相关蛋白局部黏着斑激酶(FAK)和共激活因子相关蛋白(YAP)活化情况。结果A、B两组牵拉即刻的牵张力分别增加了(22.78%±2.452%)和(15.46%±2.193%)(P<0.05)。相比B组,A组瘢痕厚度显著增加(P<0.05),胶原束粗大呈漩涡样排列紊乱,且α-SMA蛋白表达高于B组(P<0.05)。A组FAK和YAP阳性表达率均较B组增高(P<0.05;P<0.001)。结论于鼠尾3 cm处建立创面进行牵拉可以建立更接近于人增生性瘢痕的动物模型。Objective To establish an effective animal model of hypertrophic scar.Methods A 6 mm square wound was made on the SD rat tail.According to the distance from the wound to the root of rat tail,the rats were randomly divided into group A(3 cm)and group B(5 cm).The mechanical stretch device was attached on the rat tail 16 days after operation and the strain was measured.Photos were taken at 7,14,21,28 days to observe the general appearance after stress induction.After 28 days of induction,histological staining were used to measure the expression of hypertrophic scar-associated protein.The activationof stress-associated protein,FAK and YAP were also investigated.Results The immediate tension of pulling in group A and group B increased(22.78%±2.452%)and(15.46%±2.193%),respectively(P<0.05).Compared with group B,scar thickness was significantly higher in group A(P<0.05).In group A,collagen bundles were thickened and distorted and the protein expression ofα-SMA was higher than in group B(P<0.05).The activation of FAK and the positive expression rate of YAP protein in nucleus in group A were higher than that in group B(P<0.05;P<0.001).Conclusion An animal model closer to human hypertrophic scar can be established by pulling the wound at 3 cm of the rat tail.
