摘 要:目的观察杞菊汤预防PM2.5致小鼠急性肺损伤的疗效。方法将44只雄性ICR小鼠随机分为4组,正常组、模型组、乙酰半胱氨酸组和中药组。于实验第1天起,正常组、模型组小鼠灌服0.9%生理盐水,乙酰半胱氨酸组和中药组小鼠分别灌服浓度为0.0091g/ml乙酰半胱氨酸溶液和浓度为0.713g/ml的杞菊汤,每只小鼠灌服剂量为0.01ml/(g·d),连续灌服7d,于实验第8天起将正常组小鼠放入洁净仓中,其余三组小鼠放入染毒暴露仓中进行PM2.5染毒,连续放入3d,各组小鼠于实验第11天进行标本的采集和处理。用ELISA法对各组小鼠血清中TNF-α的浓度进行检测;用生化法对各组小鼠血清中MDA、GSH-PX的含量进行检测;HE染色法观察各组小鼠肺组织的病理学改变。结果与正常组比较,模型组血清中TNF-α的浓度、MDA的含量均上升,GSH-PX的含量下降,差异有统计学意义(P<0.05),与模型组比较,乙酰半胱氨酸组和中药组血清中TNF-α的浓度、MDA的含量均下降,GSH-PX的含量上升,差异有统计学意义(P<0.05)。结论PM2.5急性暴露可引起小鼠急性肺损伤,杞菊汤可通过减轻炎症反应,抑制氧化应激预防PM2.5对小鼠肺组织造成的损害。Objective To observe the efficacy of Qiju decoction in preventing acute lung injury caused by PM2.5 in mice.Methods 44 male ICR mice were randomly divided into four groups:normal group,model group,acetylcysteine group and Chinese medicine group.The experiment 1 day,mice in the normal group and model group were given 0.9%normal saline,mice in the acetylcysteine group and the Chinese medicine group were given concentration of 0.0091g/ml acetylcysteine solution and concentration of 0.713g/ml Qiju decoction respectively,each mouse was given 0.01ml/g/day,continuous given for 7days,from the 8th day of the experiment,the mice in normal group were put into the clean bin,the other three groups of mice were put into the exposure bin for PM2.5 poisoning,put it in for 3 consecutive days,samples of each group were collected and treated on the 11th day of the experiment.The concentration of TNF-αin serum was detected by ELISA,the contents of MDA and GSH-PX in serum were detected by biochemical method,HE staining was used to observe the pathological changes of lung tissues in each group of mice.Results 1.Compared with normal group,the concentrations of TNF-αand MDA were increased and the content of GSH-PX was declined in the serum of the model group,the difference was statistically significant(P<0.05),compared with model group,the concentrations of TNF-αand MDA were declined and the content of GSH-PX was increased in the serum of the acetylcysteine group and Chinese medicine group,the difference was statistically significant(P<0.05).Conclusion Acute exposure to PM2.5 can cause acute lung injury in mice,Qiju decoction can prevent lung tissue damage caused by PM2.5 in mice by reducing inflammation and inhibiting oxidative stress.
