摘 要:自2015年我国鸡群大面积暴发肝炎-心包积水综合征以来,我国鸡群鸡腺病毒流行呈现多血清型的复杂情况,甚至同一鸡群有多种血清型鸡腺病毒同时存在,其中鸡腺病毒血清4型(FAdV-4)对鸡的致病性最强。为了解FAdV-4衣壳蛋白Hexon蛋白与其他血清型鸡腺病毒抗体之间的交叉反应性,本研究将FAdV-4 HN/151025毒株的Hexon基因进行分段表达,表达蛋白分别位于CH1(1~370 aa)、CH2(371~671aa)和CH3(672~937 aa)片段。将FAdV-4 HN/151025毒株的Hexon基因的3段表达产物分别与FAdV-4、FAdV-1、FAdV-8a、FAdV-8b和FAdV-11型抗血清进行Western-blot反应。结果显示,FAdV-4 Hexon蛋白与不同血清型抗体之间均有交叉反应,但不同片段的交叉反应强度有所差异。FAdV-1型血清可与FAdV-4Hexon蛋白CH1和CH2片段发生反应,说明FAdV-4 Hexon蛋白的1~671 aa与FAdV-1型血清有交叉反应。FAdV-8a和FAdV-8b型血清仅与FAdV-4 Hexon蛋白CH2片段发生反应,说明FAdV-8a和FAdV-8b型血清与FAdV-4 Hexon蛋白的交叉反应区域在371~671 aa内。FAdV-11型血清可与FAdV-4 Hexon蛋白的3个片段发生反应,提示FAdV-11与FAdV-4型之间有交叉保护。血清4型FAdV的Hexon蛋白与不同血清型FAdV抗体间的交叉反应及其主要反应区域的研究,将对不同血清型FAdV疫苗的研制及不同血清型FAdV鉴别诊断提供依据。Hepatitis-hydropericardium syndrome(HHS)occurred in chicken flocks in China since2015.The infection of multiple serotypes of fowl adenovirus(FAdV)in chicken flocks made the disease in a complex situation.Furthermore,co-infection of multiple serotypes of FAd V in the same chicken flocks were common.FAd V-4 was the most virulent agent among the 12 serotypes of FAd V.To understand the cross-reaction the of FAd V-4 Hexon protein with the antibodies of other serotypes,the hexon protein of FAd V-4 HN/151025 strain was expressed in prokaryotic in three segments and purified,including CH1(1—370 aa),CH2(371—671 aa)and CH3(672—937 aa).Western-blot was performed to test the cross-reactivity of the expression products of the hexon gene of FAd V-4 HN/151025 with the antiserum of parental virus FAd V-4,FAd V-1,FAd V-8 a,FAd V-8 b and FAd V-11,respectively.The results showed that there were cross-reaction between FAd V-4 hexon protein and antibodies of different serotypes,but the intensity of cross-reactivity was different in some extent.The antiserum against FAd V-1 could react with CH1 and CH2 fragments of FAd V-4 hexon protein,which indicated that 1—671 aa of hexon protein has cross-reaction with FAd V-1 antiserum.The antiserum against FAd V-8 a and FAd V-8 b could only react with the CH2 fragment of FAd V-4 hexon protein,which suggested that the cross-reaction domain of FAd V-8 a and FAd V-8 b with FAd V-4 hexon protein is within 371—671 aa.There should have cross protection between FAd V-11 and FAd V-4 according to the cross reaction between the whole protein of FAd V-4 hexon with antiserum against FAd V-11.The study on cross-reaction of the FAd V-4 hexon protein with the antibodies of different serotypes of FAd V and the main reaction domain will provide the basis for the development of different serotypes of FAd V vaccine and the differential diagnosis of different serotypes of FAd V.
