摘  要:WRKY类转录因子参与植物生长发育调控及低温逆境应答。然而低温条件下WRKY参与菜心提前抽薹开花的分子机制尚不明确。本研究采用15℃低温处理菜心幼苗24 h、48 h,结果表明低温能使菜心叶片和茎尖中BcSOC1基因的表达量升高,菜心提前抽薹开花。低温处理对菜心茎尖和叶片中13个WRKY转录因子基因表达都有不同程度的影响,菜心BcWRKY22基因受低温上调表达。在拟南芥中异源表达菜心BcWRKY22基因可以使拟南芥提前抽薹开花,转基因植株叶片中AtSOC1的表达量显著高于野生型。酵母单杂交实验表明BcWRKY22转录因子可以结合BcSOC1基因的启动子。本试验说明BcWRKY22转录因子响应低温胁迫,可能通过对开花相关基因BcSOC1的启动子直接结合作用来调控菜心提前开花。The WRKY transcriptional factors were involved in the regulation of plant growth and development and respond to low temperature stress.However,the molecular mechanism of WRKY involved in early bolting and flowering in Chinese flowering cabbage under low temperature conditions is still unclear.In this study,the seedlings of Chinese flowering cabbage were treated at 15℃for 24 hours and 48 hours.The results showed that the low temperature resulted in earlier bolting,and could increase the expression levels of BcSOC1 in the leaves and shoot tips.Low temperature treatment influenced the expression of 13 WRKY genes in the shoot tips and leaves.The expression of BcWRKY22 was induced by low temperature stress.Over expression of BcWRKY22 in Arabidopsis thaliana resulted in earlier flow ering.The expression levels of AtSOC1 in the leaves of transgenic plants was significantly higher than that of wild type.Yeast one-hybrid experiments showed that BcWRKY22 could bind to the promoter of BcSOC1.These experiments demonstrated that the transcriptional factor BcWRKY22 responded to low temperature stress and may regulate the early flowering of Chinese flowering cabbage by direct binding to the promoter of the flowering-related gene BcSOC1.
