摘 要:晚期胚胎发育丰富蛋白(late embryogenesis abundant protein,LEA)普遍存在于植物中,该家族蛋白在非生物胁迫下大量积累以保护植物正常生长发育。为了探索MaLEA家族蛋白在调控香蕉非生物胁迫中的功能,本研究利用生物信息学方法对23个香蕉LEA家族的基因结构、理化性质、亚细胞定位、系统进化、表达特性及启动子进行初步研究。结果显示:MaLEA家族属于高度亲水蛋白,等电点在4.6~9.96之间,大多数定位于细胞质,同时在细胞核、叶绿体和线粒体也有分布。聚类分析发现MaLEA家族共分为7组,不同组间Motif存在一定差异;在启动子序列中,内含子数目为0~3。转录组数据显示,大多数MaLEA家族成员在香蕉果实发育的前期表达量较高;同时,MaLEA3、MaLEA5、MaLEA14和MaLEA23响应了干旱、盐和冷胁迫。进一步分析MaLEA家族基因启动子区响应元件,发现大量响应盐、干旱、高温、低温胁迫、机械损伤和茉莉酸甲酯、脱落酸激素等的顺式作用元件,推测MaLEA家族在非生物胁迫中发挥着重要作用,为深入研究香蕉MaLEA家族基因的功能提供了基础数据。The late embryogenesis abundant protein,named LEA,was greatly accumulated under abiotic stress to protect the normal growth and development of plants.In order to explore the function of MaLEA family proteins in regulating the abiotic stress.In this shudy,23 MaLEA genetic structures,physicochemical properties,subcellular localizations,phylogenies,expression characteristicses and promoter regions wereanalyzed using bioinformatics methods,based on banana genomes and transcriptome databases.The results showed that the MaLEA family proteins belonged to highly hydrophilic proteins,theoretical isoelectric points ranged from 4.60 to 9.96,most of MaLEA proteins were localized to the cytoplasm and also partly distributed in the nucleus,chloroplast and mitochondria.MaLEA proteins could be clustered into 7 groups.There were some differences in motifs between different groups.In the promoter,the number of introns was 0~3.Transcriptomic data revealed that mostMaLEA family genes showed generally high expression at the early stage of fruit development.The MaLEA3,MaLEA5,MaLEA14 and MaLEA23 genes responded to drought,salt and cold stresses.The presence of salt,dehydration,damage,high temperature,low temperature,MeJA,abscisic acid cis-acting elements in the promoters of MaLEAs suggested that MaLEAs may play important roles in abiotic stress,it provided basic data for the in-depth study of banana MaLEA family genes function.
