摘  要:为明确红宝石海棠花粉离体萌发及花粉管生长的特性,以新鲜花粉为材料,采用液体培养法,研究不同浓度的蔗糖、氯化钙、硼酸、PEG-6000及不同培养时间对其萌发率及花粉管生长的影响。结果表明,蔗糖、氯化钙、硼酸和PEG-6000均对红宝石海棠花粉萌发和花粉管生长影响显著,最适钙和硼酸浓度均为0.1 g/L,蔗糖浓度为150 g/L,PEG-6000为50 g/L,最佳培养时间为2 h,花粉萌发率达92.31%,花粉管长度达582.96μm。本试验探讨液体培养基中不同组分对红宝石海棠花粉萌发和花粉管生长的影响,筛选出红宝石海棠花粉离体萌发的最佳培养基组合,为海棠的新品种选育及理论研究提供了科学依据。In order to clear the pollen germination and pollen tube growth characteristic of Malus Royalty in vitro,the effects of the concentrations of sucrose,CaCl2,H3BO3 and PEG-6000,culture times on pollen germination of Malus spetabilis Royalty were studied using pollen liquid culture.The results showed that,culture times and the concentrations of sucrose,CaCl2,H3BO3 and PEG-6000 in the medium had significant influence on pollen germination and pollen tube growth.The optimal concentration of calcium and boric acid was 0.1 g/L,sucrose was 150 g/L,PEG-6000 was 50 g/L,and the optimal culture time was 2 h;The pollen germination rate was 92.31%,and the length of pollen tube was 582.96μm respectively.The pollen germination rate and pollen tube length were the highest at for 2 h.This experiment discussed the influence of different liquid culture mediums on pollen germination and pollen tube growth and screened out the optimum culture combination of pollen germination and pollen tube growth of Malus Royalty in vitro,and it would lay foundation of cross breeding and theoretical research of Malus Royalty.
