保肺颗粒祛痰平喘止咳作用及临床意义

摘　　要：目的研究保肺颗粒的止咳、平喘和祛痰作用及临床意义,为临床合理用药提供依据。方法通过大鼠毛细血管排痰量实验,观察其祛痰作用,取大鼠按体重随机分为空白对照组、羧甲司坦组、急支糖浆组、保肺颗粒高、中、低剂量组,连续灌胃给药7天,末次给药30 min后,麻醉后分离气管,在甲状软骨下缘正中处两软骨之间插入毛细管,观察4 h,记录每组痰液分泌量。通过对豚鼠离体气管平滑肌的影响,观察其平喘作用,取豚鼠气管环,置克-亨氏液中,待稳定后,加入终浓度1×10^-5 mol/L的氯化乙酰胆碱(Ach),至收缩高度达最大值时,加入受试物,记录加入受试物后5 min内的曲线高度,换液休息至曲线恢复至正常水平。记录各组给药前曲线高度及给药后曲线高度,计算松弛百分率。通过枸橼酸喷雾引起的豚鼠咳嗽反应,观察其止咳作用,取预选合格的豚鼠,以咳嗽次数随机分为模型对照组、氢溴酸右美沙芬组、急支糖浆组、保肺颗粒高、中、低剂量组,连续7天灌胃给药,末次给药30 min,用17.5%枸橼酸水溶液喷雾,观察记录豚鼠咳嗽潜伏期及给药后5 min内的咳嗽次数。结果对大鼠排痰量(毛细玻管法)实验表明:保肺颗粒高、中剂量能显著增加大鼠排痰量,低剂量组增加大鼠排痰量,但增加不显著。对豚鼠离体气管平滑肌的实验表明:保肺颗粒高、中、低剂量均能有效缓解乙酰胆碱引起的豚鼠离体气管平滑肌的收缩。对豚鼠的止咳(枸橼酸喷雾刺激法)实验表明:保肺颗粒高、低剂量能明显延长枸橼酸引起的豚鼠咳嗽潜伏期,能明显减少5min内咳嗽次数,潜伏期延长率有明显提高。结论保肺颗粒具有较好的祛痰、平喘、止咳作用,保肺颗粒具有较好的临床应用价值,为临床用药提供依据。Objective To study the expectorant,antiasthmatic and antitussive effects of Baofei Granules and its clinical significance in order to provide a basis for clinical rational drug use.Methods The rat capillary expectoration method was used to observe the expectorant effect.The rats were randomly divided into the blank control group,the carmastam group,Jizhi syrup group,high,medium and low dose groups of Baofei Granules according to body weight,and were administered orally for 7 days.30 minutes after the last administration,separating trachea after anesthesia,a capillary was inserted between the two cartilages in the middle of the lower edge of the thyroid cartilage,then observed for 4 hours.The sputum secretion of each group was recorded.The guinea pigs isolated tracheal smooth muscle method was used to observe the antiasthmatic effect.Taken the guinea pig tracheal ring and placed it in Gram-Henz solution.After stabilization,added acetylcholine chloride(Ach)at a final concentration of 1×10^-5 mol/L.When the contraction height reached the maximum,added the test substance and recorded the height of the curve within 5 minutes,then changed the liquid and rested until the curve returns to normal level.Before and after administration the height of the curves of each group were recorded,and the percentage of relaxation was calculated.The methods of cough induced by citric acid in guinea pigs was used to observe the antitussive effects.Pre-selected qualified guinea pigs were randomly divided into the model control group,the dextromethorphan hydrobromide group,the Jizhi syrup group,high,medium and low dose groups of Baofei Granules by cough frequency,which were administered orally for 7 days.30 minutes after the last administration,sprayed with a 17.5%citric acid aqueous solution,then the cough latency of guinea pigs and the number of coughs within 5 minutes were observed and recorded.Results Experiments on rat sputum output(capillary tube method)showed the high and medium dose groups of Baofei Granules could promote expect
