摘  要:以虎头兰组培苗为试材,采用单因素和正交实验设计,研究了添加不同种类激素和含量对类原球茎诱导的影响以及不同培养基类型、不同激素含量、活性炭含量对类原球茎增殖的影响,并对诱导出的类原球茎进行包埋保存,以期为虎头兰的组培繁殖和种质保存提供新的方法。结果表明:无菌苗诱导类原球茎添加3mg·L^-1 TDZ最佳,30d诱导率到达80%,90d达到100%;类原球茎增殖的最佳培养基为花宝1号3g·L^-1+0.1mg·L^-1 NAA+3mg·L^-16-BA+1g·L^-1活性炭,增殖系数可达4.03;类原球茎通过3.5%海藻酸钠溶液和0.1mol·L^-1 CaCl2包埋在4℃黑暗条件下保存120d,萌发率可以达到48%,植株再生率可达44%。In order to provide a new method for the propagation and germplasm resources conservation of Cymbidium hookerianum,the tissue culture seedlings of Cymbidium hookerianum were employed to evaluate the effects of adding different kinds and content of hormone on the induction of protocormlike bodys(PLBs),the effects of different media types,different hormone contents and activated carbon(AC)content on PLBs propagation,and the induced PLBs preserved via encapsulating by using single factor and orthogonal designs.The results showed that the addition of 3mg·L^-1 TDZ was optimal to the induction of PLBs;the optimal medium for PLBs propagation was 3 g·L^-1 Hyponex 1+0.1mg·L^-1 NAA+3mg·L^-16-BA+1g·L^-1 AC;PLBs encapsulated by 3.5%sodium alginate solution and 0.1 mol·L^-1 CaCl2 stored at 4℃in the dark for 120days,the germination rate reached 48%,conversion rate reached 44%.
