摘 要:目的建立一种固相萃取(SPE)-超高液相色谱-串联质谱(UPLC-MS/MS)的方法,测定儿童尿液中的5种邻苯二甲酸酯代谢物。方法选取上海市某小学8~11岁的健康学生,取尿样采用OASIS Max SPE小柱净化富集后,经苯基柱分离,在多反应监测模式(MRM)下使用UPLC-MS/MS进行分析。结果5种代谢物在0.1~200.0 ng/mL线性范围内良好,相关系数均>0.99,检出限在0.01~0.14 ng/mL之间,样品平均加标回收率在85.68%~111.21%之间,相对标准偏差(RSD)均<11.00%。结论该方法操作简便、回收率较高、重现性好且检出限低,适用于儿童尿液中5种邻苯二甲酸代谢物的含量测定。Objective To develop a method for determination of five phthalate metabolites in children urine by solid phase extraction(SPE)-ultraperformance liquid chromatography-tandem mass spectrome try(UPLC-MS/MS).Methods The healthy students aged 8-11 years old were selected from a primary school in Shanghai,to collect their urine samples.Phthalate metabolites were extracted from urine samples with OASIS Max SPE column,and then separated on a CSH column,and analyzed by UPLC-MS/MS under Multi reaction monitoring mode(MRM).Results Good linear relationships were obtained in the range of 0.1-200.0 ng/mL with the correlation coefficients above 0.99 for all tested five metabolites.The limits of detection were 0.01-0.14 ng/mL.The recoveries were in the range of 85.68%-111.21%,and all relative standard deviations(RSD)were lower than 11.00%.Conclusion This method had the advantages of easy operation,high recovery,good reproducibility and low detection limit,and it is suitable for detection of five phthalate metabolites in children urine.
