摘 要:目的探讨胆固醇调节元件结合蛋白(SREBP)的裂解激活蛋白(SCAP)超表达对人类肾系膜细胞(HMCs)脂代谢的影响及其分子机制。方法分别将质粒pcDNA3.1(非超表达对照组)和pcDNA-SM22-SCAP(观察组)转染入HMCs,在含和不含低密度脂蛋白(LDL)的培养基中进行孵育。采用油红O染色和细胞内胆固醇测定分别检测不同条件下HMCs的胞内胆固醇含量。用realtime PCR和western blot分别检测SCAP、羟甲基戊二酸单酰辅酶A(HMG-CoA)还原酶、LDL受体(LDLr)的mRNA和蛋白的表达,及核蛋白SREBP2 N末端(N-SREBP2)的蛋白表达。结果相同培养基条件下,观察组的细胞内胆固醇含量明显高于对照组。对照组细胞加入LDL后,HMG-CoA还原酶、LDLr的mRNA和蛋白以及N-SREBP2蛋白表达明显下调;而观察组细胞不论是否加入LDL,上述基因mRNA和蛋白的表达均不下调。结论 SCAP超表达破坏了HMCs中LDL受体途径正常的生理调节机制,在脂质引发的肾损害中有重要作用。Objective To explore the effect of SCAP overexpression on lipid metabolism of human mesangial cells(HMCs), and its molecular mechanism. Methods pcDNA3.1(the control group) and pcDNA-SM22-SCAP(the observation group) were transfected into HMCs respectively, and the cells were incubated with and without low density lipoprotein(LDL). The cholesterol content in HMCs at different conditions were examined by Oil red O staining and intracellular cholesterol measurement. The expression of SCAP, HMG-CoA reductase, mRNA and N-SREBP2 were examined by real-time PCR and western blot. Results In the same medium, the cholesterol content in HMCs in the observation group was obviously higher than that of the control group. After adding LDL in the cells of the control group, the expression of HMG-CoA reductase, mRNA and N-SREBP2 obviously reduced. However, whether adding LDL in the cells of the observation group or not, the expression didn’t reduce. Conclusion The SCAP overexpression damage physiological regulatory mechanism of LDL receptors in HMCs and play an important role in lipid-mediated renal injury.
