摘 要:脱细胞支架通常被用于辅助组织再生修复,然而脱细胞支架植入后与宿主组织间的相互作用和植入转归机制目前尚不清楚。为此本研究制备了牛心包脱细胞支架(S1)和牛皮脱细胞支架(S2)两种材料,通过体外性能测定和体内动物试验,研究脱细胞支架植入后与宿主组织之间的相互作用和其植入转归过程。试验结果显示,两种支架材料植入1个月后均能够与比格犬自体硬脑膜组织融合生长,但是S2植入3个月被完全降解,而硬脑膜缺损位置未见新生硬脑膜组织生成,说明脱细胞支架与宿主组织之间成功整合并不能保证组织再生修复。脱细胞支架植入后会重启ECM动态变化过程,脱细胞支架恢复ECM动态平衡是其能否发挥组织再生/修复能力的必要条件。Acellular scaffold is usually used to assist tissue repair and regeneration.However,many problems remained.For instance,the interactions between acellular scaffolds and host tissues and the mechanism of tissue repairing and regeneration.Therefore,two kinds of acellular scaffolds were prepared,including sample 1(S1,decellularized bovine pericardium)and sample 2(S2,decellularized bovine skin).The in vitro characteristics and in vivo performance in Beagles’model were measured.The results showed that both of the two scaffolds were successfully integrated with beagles’autologous dura after one month implantation.S1 remained its original shape after 3 months’implantation,However,S2 were completely degraded after 3 months implantation.There were no newly regenerated dura tissues.The result indicated that successful integration could not guarantee tissue regeneration.The ECM dynamics would restart after surgical implantation.The ECM dynamics balance in acellular scaffolds should also be established.The restoration of ECM dynamics in acellular scaffold was necessary for tissue repairing/regeneration.
