连花清咳片对大鼠急性支气管炎模型的保护作用及机制研究

摘　　要：目的研究连花清咳片对细菌合并烟熏所致的大鼠急性支气管炎的保护作用及机制。方法建立混合烟熏法同时经鼻滴入卡他莫拉菌复制大鼠急性支气管炎模型。实验分为正常对照组,模型对照组,银黄清肺胶囊对照药组(0.18 g·kg^-1)及连花清咳片低、中、高剂量组(1.47、2.93和5.86 g生药·kg^-1)。给药组按10 mL·kg^-1灌胃给予相应药液,1次·天^-1,连续14 d。观察各组大鼠的一般状态;ELISA检测肺泡灌洗液中IL-6、IL-8、IL-10、TNF-α;比色法检测NO、SOD、MDA;流式细胞仪检测肺泡灌洗液中CD4^+/CD3^+、CD8^+/CD3^+细胞的比例,计算CD4^+/CD8^+细胞的比例;HE染色观察支气管及肺组织病理学改变。结果与正常对照组比较,模型对照组大鼠肺脏中炎症因子IL-6、IL-8、TNF-α水平增加,IL-10水平降低,MDA含量增加,NO、SOD含量降低,CD4^+/CD8^+细胞比值降低;与模型对照组比较,连花清咳片各剂量组大鼠肺脏中炎症因子IL-6、IL-8、TNF-α的表达减少,IL-10的表达增加,MDA含量减少,NO、SOD的含量升高,CD4^+/CD3^+、CD8^+/CD3^+细胞的比例以及CD4^+/CD8^+细胞的比值增加。连花清咳片中、高剂量组和银黄清肺胶囊对照药组动物支气管及肺脏病理改变均有明显改善。结论连花清咳片对细菌合并烟熏所致的大鼠急性支气管炎具有显著的保护作用,其机制与降低肺脏组织中炎性因子,抗氧化和提高细胞免疫功能有关。Objective To determine the protective effect and mechanism of Lianhua Qingke tablets on acute bronchitis in rats induced by bacteria and smoking.Methods Rat models of acute bronchitis induced by bacteria and smoking were established.The rats were divided into a normal control group,a model control group,a Yinhuang Qingfei capsule control group(0.18 g·kg^-1),3 dose groups of Lianhua Qingke tablets(1.47,2.93,and 5.86 g dried medicinal herbs·kg^-1),once daily for 14 days.The general state of the rats in each group was observed.ELISA was used to detect IL-6,IL-8,IL-10,and TNF-α,and colorimetric method was used to detect NO,SOD and MDA in the alveolar lavage fluid.Flow cytometry was used to detect the proportion of CD4^+/CD3^+and CD8^+/CD3^+cells in the alveolar lavage fluid,and then calculated the proportion of CD4^+/CD8^+cells.HE staining was used to observe the pathological manifestation.Results Compared with the normal control group,the expression of inflammatory factors IL-6,IL-8 and TNF-αwas increased in the lungs of the model group,the expression of IL-10 was decreased,the content of MDA was increased,the content of NO and SOD was decreased,and the proportion of CD4^+/CD8^+cells was decreased.Compared with the model control group,the expression levels of inflammatory cytokines IL-6,IL-8 and TNF-αin the lungs of rats in all Lianhua Qingke groups were decreased,the expression of IL-10 was increased,the content of MDA decreased,the content of NO and SOD was increased,and the proportion of CD4^+/CD3^+,CD8^+/CD3^+,and CD4^+/CD8^+cells was increased.The pathological changes of bronchi and lungs in Lianhua Qingke tablet groups and Yinhuang Qingfei control group were significantly improved.Conclusion Lianhua Qingke tablets have protective effect on acute bronchitis in rats induced by bacteria and smoking,which is related to the reduction of inflammatory factors in the lung tissue,antioxidant effect and improvement of cellular immune function.
