摘 要:为了解安徽省规模化猪场贾第虫感染情况及分子特性,从安徽省多地共采集500份新鲜猪粪样,提取基因组DNA,首先采用基于蓝氏贾第虫SSUrRNA基因套式PCR对所有粪便进行检测,并对获得的阳性样本分别基于贾第虫TPI和GDH基因的PCR扩增,然后对获得的PCR产物进行测序和分析,确定贾第虫的聚集体。结果显示,500份猪粪便样品中,SSUrRNA基因套式PCR仅在利辛猪场检测出1例贾第虫阳性样本,猪贾第虫阳性率为0.20%。TPI和GDH基因的测序分析显示该贾第虫基因型为聚集体E。To determine the prevalence and molecular characteristics of the isolates of G.duodenalis in Anhui province,500 fecal samples were detected by PCR targeting of SSUrRNA gene for the presence of G.duodenalis after fecal samples were collected and genomic DNA was extracted.Subsequently a nested-PCR targetingTPI gene and a hemi-nested-PCR targeting GDH gene were performed in the SSUrRNA-positive samples.The obtained PCR products were sequenced and bioinformatically analyzed to identify the assemblage of G.duodenalis.Of the 500 pig feces samples,only 1 positive sample of G.duodenalis from the pig farm in Lixin was detected by nested PCR of SSUrRNA gene,and the positive rate of G.duodenalis in pigs in Anhui province was 0.20%.Sequencing analysis of TPI and GDH genes showed that the genotype of the isolate obtained in this study was assemblage E.
