摘 要:为了探讨人参皂苷Rg1对心衰大鼠心功能的保护作用,并对其作用机制进行研究,建立100只雄性大鼠心衰模型并分为模型组,阳性药组给予依那普利[15 mg/(kg·d)],人参皂苷Rg1低[1 mg/(kg·d)]、中[2 mg/(kg·d)]和高剂量组[4 mg/(kg·d)],20例/组。检测大鼠左室收缩末期内径(Left ventricular end-systolic diameter,LVEDV)左室舒张末期内径(Left ventricular end-diastolic diameter,LVESV)、左室射血分数(Ejection fraction,EF)、左心室最大内压上升/下降速率(Maximum left ventricular pressure rise/fall rate,±dp/dtmax)、心肌细胞Na^+-K^+ATP酶、Ca^2+-Mg^2+-ATP酶和总ATP(Total ATP,T-ATP)酶、总糖原、碱性磷酸酶(Alkaline phosphatase,ACP)、乳酸脱氢酶(Lactate dehydrogenase,LDH)及心肌细胞胞浆内心肌肌浆网钙离子ATP酶(Sarcoplasmic reticulum Ca^2+-ATPase,SERCA2a)的表达。5组大鼠的心脏EF、LVEDV、LVESV、±dp/dtmax水平存在显著性差异(P<0.05);与模型组、人参皂苷Rg1低剂量、中剂量组相比,高剂量组EF、±dp/dtmax明显增高(P<0.05),LVEDV、LVESV明显降低(P<0.05)。5组大鼠心肌细胞的Na^+-K^+-ATP酶、Ca^2+-Mg^2+-ATP酶、T-ATP酶、总糖原、ACP、LDH的含量存在显著性差异(P<0.05);与模型组、人参皂苷Rg1低剂量、中剂量组相比,高剂量组Ca^2+-Mg^2+-ATP酶、T-ATP酶、总糖原、ACP、LDH明显增高(P<0.05),Na^+-K^+-ATP酶明显降低(P<0.05)。5组大鼠SERCA2a的表达存在显著性差异(P<0.05),与模型组、人参皂苷Rg1低剂量、中剂量组相比,高剂量组SERCA2a的表达明显增高(P<0.05)。EF、±dp/dtmax、Ca^2+-Mg^2+-ATP酶、T-ATP酶、总糖原、ACP、LDH、SERCA2a与人参皂苷Rg1剂量呈正相关(P<0.05);LVEDV、LVESV、Na^+-K^+-ATP酶与人参皂苷Rg1剂量呈负相关(P<0.05)。人参皂苷Rg1可改善心衰大鼠的心功能,与增高EF、±dp/dtmax、Ca^2+-Mg^2+-ATP酶、T-ATP酶、总糖原、ACP、LDH、SERCA2a及降低LVEDV、LVESV、Na^+-K^+-ATP酶有关,且呈剂量依赖性相关。To investigate the protective effect of ginsenoside Rg1 on cardiac function,as well as its possible mechanism in rats with heart failure,a total 100 male rats with heart failure were established and divided into 5 groups:the model groups,the positive drug group[enalapril(15 mg/(kg·d))],Rg1 low group(1 mg/(kg·d)),Rg1 medium-dose group(2 mg/(kg·d))and Rg1 high-dose group(4 mg/(kg·d)),20 rats per group.The left ventricular end-systolic diameter(LVEDV),left ventricular end-diastolic diameter(LVESV),left ventricular ejection fraction(EF),maximum left ventricular pressure rise/fall rate(±dp/dtmax),myocardial cells Na^+-K^+-ATP,Ca^2+-Mg^2+-ATP and total ATP(T-ATP)enzyme,total glycogen,alkaline phosphatase(ACP),lactate dehydrogenase(LDH)and sarcoplasmic reticulum Ca^2+-ATPase(SERCA2 a)in myocardial cells were observed.The cardiac EF,LVEDV,LVESV,and±dp/dtmaxlevels of 5 groups of rats were significantly different(P<0.05).Compared with the model group,the ginsenoside Rg1 low-and medium-dose groups,the EF,±dp/dtmaxsignificantly increased(P<0.05),and LVEDV and LVESV significantly decreased(P<0.05).The contents of Na^+-K^+-ATP,Ca^2+-Mg^2+-ATP,T-ATPase,total glycogen,ACP,LDH in rat cardiomyocytes of 5 groups were significantly different(P<0.05).Compared with the model group,ginseng Rg1 low-and medium-dose groups,the Ca^2+-Mg^2+-ATP,T-ATPase,total glycogen,ACP,and LDH in the ginseng Rg1 high-dose group significantly increased(P<0.05),and Na^+-K^+-ATP significantly reduced(P<0.05).There were significant differences in the expression of SERCA2 a in5 groups(P<0.05).Compared with the model group,ginseng Rg1 low-and medium-dose groups,the expression of SERCA2 a in the ginseng Rg1 high-dose group were significantly higher(P<0.05).EF,±dp/dtmax,Ca^2+-Mg^2+-ATP,T-ATPase,total glycogen,ACP,LDH,SERCA2 a and ginsenoside Rg1 dose were positively correlated(P<0.05).The LVEDV,LVESV,Na^+-K^+-ATP were negatively correlated with ginsenoside Rg1 dose(P<0.05).Ginsenoside Rg1 could improve heart function in rats with heart failure,and increa
