摘  要:自体皮片移植是治疗烧伤、创伤及其他因素所致皮肤缺损的有效方法。将富血小板纤维蛋白(PRF)膜应用于犬自体皮片移植并进行术后评价,以探讨PRF在犬自体皮片移植中的应用效果。试验采取犬自体静脉血制备PRF膜标本,并将其应用于犬自体皮片移植试验,术部选在犬双侧前肢胫骨前缘处皮肤,采用自体对照的试验方法,左前肢为试验组,右前肢为空白组,在术后第3、5、7、14、28天分别对移植部位进行局部观察,HE染色镜检,测算PDGF-BB及TGF-β1表达阳性率。结果30组移植皮片均成活,同时PRF提高了PDGF-BB与TGF-β1的表达水平,有助于内皮细胞、巨噬细胞、成纤维细胞和角质细胞增殖分化、加速移植创面的愈合。术后评价证实PRF在犬自体皮片移植术中对止血、控制炎症效果显著,能够缩短创口愈合时间提高创口愈合率。Autologous skin transplantation is an effective method to treat skin defects caused by burn,trauma and other factors.In this study,the platelet-rich fibrin(PRF)membrane was applied to canine autologous reticular skin graft and postoperative evaluation to explore the clinical effect of PRF in canine autologous reticular skin graft transplantation.In this experiment,the PRF structure is observed through a microscope after the PRF membrane specimen was prepared by canine autologous venous blood,and it was applied to the canine autologous reticular skin graft test.The operation was performed on the skin of the anterior border of canine's bilateral forelimbs tibia,using the self-controlled test method,the left side was the experimental group,and the right side was the blank group.The results showed that the 30 groups of transplanted skin grafts survived.Local observation and HE staining were performed on the graft site at 3,5,7,14 and 28 days after surgery,respectively,and calculated the positive expression rates of PDGF-BB and TGF-β1.Postoperative evaluation confirmed that PRF can significantly reduce hemostasis and control inflammation in canine autologous reticular skin graft,which can shorten wound healing time and increase wound healing rate.At the same time,PRF can not only improve the expression of PDGF-BB and TGF-β1,also promote the proliferation and differentiation of endothelial cells,macrophages,fibroblasts and keratinocytes,which helps accelerate the healing of transplanted wounds.
