摘 要:目的研究氧化槐果碱(oxysophocarpine,OSC)对脑缺血再灌注大鼠氧化损伤和炎性反应的缓解作用。方法采取线栓法建立局灶性脑缺血再灌注模型,造模后24 h后分别灌胃给药OCS 0.25、1、4 mg/kg,给药10 d后,比较各组大鼠的神经功能评分、脑组织形态结构、氧化应激指标、炎症指标、凋亡相关蛋白、Nrf2/HO-1/NQO1通路及NF-κB蛋白表达水平。结果OSC干预后,模型大鼠的神经功能评分、MDA、LDH、IL-6、IL-1β、Caspase-3、Bax表达水平、NF-κB表达水平均明显降低(P<0.05),而SOD、IL-10、IL-4、Bcl-2、Nrf2、HO-1、NQO1表达水平较显著升高(P<0.05)。结论OSC可能通过上调Nrf2/HO-1/NQO1通路和抑制NF-κB的活化缓解脑缺血再灌注引起氧化损伤和炎性反应。Objective To study the alleviating effects of oxysophocarpine(OSC)on oxidative damages and inflammatory responses in rats with cerebral ischemia-reperfusion.Methods The focal cerebral ischemia-reperfusion models were established by suture-occluded method.Twenty-four hours after modeling,0.25,1 and 4 mg/kg OCS were intragastrically administered,respectively.After 10-day administration,the scores of neurological function,morphology of brain tissues,levels of oxidative stress indexes,inflammation indexes,apoptosis-related proteins,Nrf2/HO-1/NQO1 pathway and NF-κB protein were compared bwteen groups.Results After OSC intervention,the scores of neurological function,and the levels of MDA,LDH,IL-6,IL-1β,caspase-3,Bax and NF-κB were significantly decreased(P<0.05),while the expression levels of SOD,IL-10,IL-4,Bcl-2,Nrf2,HO-1 and NQO1 were significantly increased in model rats(P<0.05).Conclusion OSC can alleviate oxidative damages and inflammatory responses caused by cerebral ischemia-reperfusion by up-regulating Nrf2/HO-1/NQO1 pathway and inhibiting NF-κB activation.
