摘  要:目的验证亚甲蓝光动力疗法(MB-PDT)对红色毛癣菌的体外抑制作用;探索不同浓度亚甲蓝(MB)和不同光照能量密度介导的光动力疗法(PDT)对红色毛癣菌生物活性的影响,为临床治疗红色毛癣菌感染提供理论依据。方法参考CLSI M38-A方案,确定MB-PDT抑制红色毛癣菌的MIC值;在96孔板培养红色毛癣菌,用MTT法检测在不同MB浓度、不同光照能量密度下MB-PDT对红色毛癣菌生物活性的影响。结果红光能量密度68 J/cm2时,MB-PDT体外抑制红色毛癣菌的MIC值为6μg/mL;光照能量密度为60 J/cm2时,MB-PDT对红色毛癣菌的抑制作用随MB浓度的升高逐渐增加,MB浓度增加至10μg/mL时,抑制作用达到最大;继续增加MB浓度至20μg/mL,红色毛癣菌抑制作用无明显增加。MB浓度为10μg/mL时,随着光照能量密度的增加MB-PDT抑菌效应逐渐增强,当光照能量密度为34 J/cm2时,其红色毛癣菌抑制作用达到最大。结论MB-PDT在体外可以有效抑制红色毛癣菌;在一定范围内,MB-PDT对红色毛癣菌的抑制作用与MB浓度与光照能量密度呈正相关。Objective To test the effectiveness of methylene blue mediated photodynamic therapy(MB-PDT)in treating Trichophyton rubrum(T.rubrum);the influence of concentration of MB and energy density on the effects of MB-PDT on T.rubrum was also assessed.To provide theoretical basis for treatment of T.rubrum infection.Methods The minimum inhibitory concentration(MIC)values were determined by the Clinical and Laboratory Standards Association(CLSI)M38-A protocol. T.rubrum was cultured in a 96-well plate,and the effects of different concentrations of MB and different light energy density mediated MB-PDT on T.rubrum were detected by MTT assay,when the light wavelength was fixed on(632±10)nm.Results When the energy density of red light was 68 J/cm2,the MIC value of MB-PDT inhibiting T.rubrum in vitro was 6 μg/mL.When the light energy density was 60 J/cm2,the inhibitory effect of MB-PDT on T.rubrum increased gradually with the increase of MB concentration.When the MB concentration increased to 10 μg/mL,the inhibitory effect reached the maximum.The inhibition of T.rubrum was not significantly increased by increasing the MB concentration to 20 μg/mL.When MB concentration was 10 μg/mL,the inhibitory effect of MB-PDT gradually increased with the increase of light energy density.When the light energy density was 34 J/cm2,the inhibitory effect of MB-PDT reached the maximum.Conclusion MB-PDT has the inhibitory function on T.rubrum in vitro,which is positively correlated with MB concentration and light energy.
