摘  要:目的探讨三维适形调强放疗(intensity-modulated radiation therapy,IMRT)的放射线对鼠骨包虫生发细胞的细胞周期及凋亡因子caspase-9的影响。方法骨包虫病子午沙鼠20只,将鼠骨包虫内囊中的生发细胞分离并进行体外培养,将体外培养成功的骨包虫生发细胞分为IMRT组和对照组。IMRT组给予总剂量为40 Gy的IMRT放疗,对照组给予假照射。放疗结束24 h后,采用流式细胞仪检测2组骨包虫生发细胞的细胞周期,采用ELISA法检测骨包虫生发细胞中凋亡相关蛋白caspase-9蛋白水平,采用实时荧光定量PCR法检测骨包虫生发细胞caspase-9 mRNA相对表达量。结果放疗结束24 h后,IMRT组骨包虫生发细胞S期细胞比率[(65.65±7.46)%]明显高于对照组[(32.49±4.23)%],G0/G1期[(32.37±4.94)%]和G2/M期[(1.98±0.19)%]细胞比率明显低于对照组[(53.95±4.55)%、(13.55±2.48)%](P<0.05)。IMRT组生发细胞caspase-9蛋白水平[(0.071±0.012)pmol/L]和caspase-9 mRNA相对表达量(30.21±6.99)明显高于对照组[(0.011±0.004)pmol/L、12.17±3.01](P<0.05)。结论IMRT可改变鼠骨包虫生发细胞的细胞周期,抑制生发细胞的增殖,可能是通过调节凋亡相关蛋白caspase-9水平而实现。Objective To investigate the effects of intensity-modulated radiation therapy(IMRT)on the cell cycle of germinal cells of bone hydatid and caspase-9 in Meriones meridianuses.Methods The germinal cells of bone echinococcosis were separated and cultured in vitro in 20 Meriones meridianuses with bone echinococcosis.The successfully cultured germinal cells of bone hydatid were divided into IMRT group and control group.IMRT group received IMRT at a total dose of 40 Gy,and control group received no IMRT.The cell cycle of germinal cells of bone hydatid was detected by flow cytometry,the level of caspase-9 protein was detected by ELISA,and the relative expression of caspase-9 mRNA was detected by real-time fluorescence quantitative PCR in 24 h after IMRT.Results In 24 h after IMRT,the ratio of germinal cells of bone hydatid in S phase was higher in IMRT group((65.65±7.46)%)than that in control group((32.49±4.23)%),while the ratios of germinal cells in G0/G1 phase and G2/M phase were lower in IMRT group((32.37±4.94)%,(1.98±0.19)%)than those in control group((53.95±4.55)%,(13.55±2.48)%)(P<0.05).The level of caspase-9 protein and the relative expression of caspase-9 mRNA were higher in IMRT group((0.071±0.012)pmol/L,30.21±6.99)than those in control group((0.011±0.004)pmol/L,12.17±3.01)(P<0.05).Conclusion IMRT can change the cell cycle and suppress the proliferation the germinal cells in Meriones meridianuses with bone echinococcosis,probably by regulating the level of caspase-9 protein.
