摘  要:目的研究人黄体生成素/人绒毛膜促性腺激素受体(LHCGR)基因rs10495960和rs7579411位点多态性在广西人群中的分布特征,比较不同种族人群的分布性差异。方法本实验采用DNA测序法和多重单碱基延伸PCR(SNaPshot),对573例右江民族医学院附属医院生殖中心门诊健康体检就诊者LHCGR基因的rs10495960和rs7579411位点基因型进行检测,与国际人类基因组单体型图计划(HapMap)公布的北京人群(HapMap-CHB)、日本人群(HapMap-JPT)、欧洲人群(HapMap-CEU)及非洲人群(HapMap-YRI)LHCGR基因多态性数据进行比较。结果广西人群中,rs10495960位点包含GG、GA、AA基因型,以G/A基因型较常见(46.07%),等位基因以G较常见(64.05%);rs7579411位点包含CC、CT、TT基因型,以C/T基因型较常见(47.12%),等位基因以C较常见(62.65%)。其中rs10495960位点各基因型和等位基因频率在男、女两组间分布差异都有统计学意义(P<0.05);rs7579411位点各基因型和等位基因频率在男、女两组间分布差异均无统计学意义(P>0.05);广西地区人群rs10495960位点的基因型频率与HapMap-JPT、HapMap-CEU、HapMap-YRI比较,差异均有统计学意义(P<0.05),与HapMap-CHB比较,差异无统计学意义(P>0.05);rs10495960位点的等位基因频率与HapMap-CHB、HapMap-JPT、HapMap-CEU、HapMap-YRI比较,差异均有统计学意义(P<0.05)。广西地区人群rs7579411位点的基因型频率与HapMap-JPT、HapMap-YRI比较,差异均有统计学意义(P<0.05),与HapMap-CHB、HapMap-CEU比较,差异无统计学意义(P>0.05);rs7579411位点的等位基因频率与HapMap-JPT、HapMap-YRI比较,差异均有统计学意义(P<0.05),与HapMap-CHB、HapMap-CEU比较,差异无统计学意义(P>0.05)。结论广西人群中LHCGR基因的rs10495960位点多态性在男女中的分布存在差异性,rs10495960和rs7579411位点多态性与其他种族间存在差异。Objective To study the distribution characteristics of luteinizing hormone/choriogonadotropin receptor(LHCGR)gene rs10495960 and rs7579411 loci polymorphisms in Guangxi population and compare the distribution differences among different ethnic groups.Methods In this study,DNA Sequencing method and Multiple Single-base Extended PCR(SNaPshot)were used to detect genotypes of LHCGR gene at the rs10495960 and rs7579411 loci in 573 patients who underwent out-patient physical examination at the Reproductive Center of Affiliated Hospital of Youjiang Medical University for Nationalities,and the results were compared with the LHCGR gene polymorphism data of HapMap-CHB,HapMap-JPT,HapMap-CEU and HapMap-YRI published by the International HapMap Project(HapMap).Results In Guangxi population,rs10495960 loci contained genotypes of GG,GA and AA,among which G/A were more common(46.07%),and allele G was more common(64.05%);rs7579411 loci contained genotypes of CC,CT and TT,among which C/T were more common(47.12%),and allele C was more common(62.65%).The distribution differences in genotypes and alleles frequencies at rs10495960 loci between male and female group were all statistically significant(P<0.05),while the distribution differences in genotypes and alleles frequencies at rs7579411 loci between male and female group were all not statistically significant(P>0.05).When the genotype frequency at rs10495960 loci in Guangxi population was compared with those of HapMap-JPT,HapMap-CEU and HapMap-YRI,the differences were all statistically significant(P<0.05),but the difference was not statistically significant when compared with HapMap-CHB(P>0.05).When the allele frequency at rs10495960 loci in Guangxi population was compared with that of HapMap-CHB,HapMap-JPT,HapMap-CEU and HapMap-YRI,the differences were all statistically significant(P<0.05).When the genotype frequency at rs7579411 loci in Guangxi population was compared with those of HapMap-JPT and HapMap-YRI,the differences were all statistically significant(P<0.05),but the diffe
