紫薯组培快速繁殖技术初探

摘　　要：本研究以欣薯2号紫薯催芽得到的顶芽及带腋芽的茎段作为外植体,初步探究外植体的消毒处理方式,诱导培养、增殖培养、生根培养中不同激素配比对外植体生长发育的影响,以期为紫薯的快速繁殖提供参考。初步试验结果表明,欣薯2号紫薯在MS+6-BA 0.75 mg·L-1+NAA 0.1 mg·L-1+3.0%蔗糖+0.7%琼脂诱导培养基上出芽率最高,在MS+6-BA 3 mg·L-1+NAA 0.2 mg·L-1+3.0%蔗糖+0.7%琼脂增殖培养基上增殖倍数最高。在生根培养中对不同浓度的IBA和NAA分别探究发现NAA 0.4 mg·L-1更有利于组培苗的生根。Xinshu 2 purple sweet potato was selected as the material,axillary buds and stems with the purple potato germination were used as explants for primary culture,subculture,the second subculture,rooting culture.I had studied the treatment of disinfection of explant,and explored the influence on the growth and development of explants in the ratio of different hormone during the primary culture,subculture,and rooting culture,in order to provide reference for the rapid propagation of purple sweet potato.The primary results show that the germination percentage of Xinshu 2 purple sweet potato on induction medium of the MS+6-BA 1 mg·L-1+NAA 0.1 mg·L-1+3.0%sucrose+0.7%agar is the highest.And the proliferation coefficient on the medium of MS+6-BA 3 mg·L-1+NAA 0.2 mg·L-1+3.0%sucrose+0.7%agar is the highest.In the rooting culture of different concentrations of IBA and NAA,it is found that the 0.4 mg/L of NAA on rooting be better to sterile buds.
