摘  要:为探究泡桐丛枝病(Paulownia witches’broom,PaWB)的表观遗传机制,以白花泡桐健康苗(PF)和丛枝病苗(PFI)为材料,采用全基因组甲基化测序(WGBS)技术对白花泡桐丛枝病发生过程中全基因组DNA甲基化进行系统研究。结果表明,白花泡桐体内的DNA甲基化主要有mCG、mCHH和mCHG 3种类型;白花泡桐被植原体侵染后,C位点发生甲基化的比率下降,且发病后CHH的甲基化比率降低而CG和CHG的甲基化比率升高。此外,DNA甲基化主要发生在CG序列;差异甲基化区域(DMR)鉴定研究中共检测到了109334个DMR,其中白花泡桐患病后高甲基化DMR和低甲基化DMR分别有37408个和71926个;通过DNA甲基化和转录组的差异表达基因关联分析,找到了6个与PaWB发生相关的差异甲基化基因(DMG),功能分别涉及到植物防御反应、光合作用、植物病原互作、植物激素信号转导等生物学过程。In order to explore the epigenetic mechanism of Paulownia witches’broom(PaWB),using Paulownia fortunei seedings(PF)and phytoplasma-infected Paulownia fortunei seedings(PFI)as the material,the Whole Genome Bisulfite Sequencing(WGBS)technology was used to study the DNA methylation of the whole genome systematically during the occurrence of PaWB.The results showed that there were three types of DNA methylation(mCG,mCHH,and mCHG)in PF.The ratio of methylation at the C site decreased after the phytoplasma infection of PF,and the methylation ratio of CHH reduced,while the methylation ratio of CG and CHG increased.In addition,DNA methylation mainly occurred in the CG sequence.A total of 109,334 DMR were detected in differential methylated region(DMR),of which 37,408 and 71,926 were hypermethylated DMR(hyper-DMR)and hypomethylated DMR(hypo-DMR),respectively.By association analysis of differentially expressed genes through DNA methylation and transcriptome,six differentially methylated genes(DMG)related to the occurrence of PaWB were found,whose functions are related to biological processes such as plant defense response,photosynthesis,plant pathogen interaction,and plant hormone signal transduction.
