桂参通脉胶囊对心衰大鼠心室重构MMP-1及TIMP-1的表达研究

摘　　要：目的:研究桂参通脉胶囊对心衰大鼠心室重构MMP-1以及TIMP-1表达。方法:腹腔注射盐酸阿霉素(ADR)制备大鼠心衰模型。将60只成年雄性SD大鼠随机分为正常对照组、心衰模型组和桂参通脉低、中、高剂量组及卡托普利组,每组10只,干预4周后,提取小鼠心肌组织,采用蛋白免疫印迹法,以凝胶成像系统分析MMP-1及TIMP-1的水平。结果:MMP-1在心衰模型组的表达程度显著升高,采用桂参通脉胶囊干预后发现,中、高剂量组可显著降低MMP-1蛋白表达,且明显低于卡托普利组(P<0.05)。TIMP-1的蛋白表达在正常组最高,心衰组表达最低,通过药物干预后发现,卡托普利组、桂参通脉中、高剂量组均能上调TIMP-1的表达,其中以桂参通脉高剂量组上调最明显(P<0.05)。结论:桂参通脉胶囊能降低CHF大鼠MMP-1水平,提高TIMP-1的表达,降低MMP-1/TIMP-1比值,延缓CHF发展,逆转心室重构。Objective:To study the effect of Guishen Tongmai Capsule on the expressions of MMP-1 and TIMP-1 in heart failure rats with ventricular remodeling.Methods:The heart failure model was established by intraperitoneal injection of Adriamycin hydrochloride(ADR).A total of 60 SD rats were randomly divided into the normal group,the model group,the Guishen Tongmai Capsule groups of low-dose,medium-dose and high-dose,and the Captopril group,with 10 rats in each group.After 4 weeks of intervention,the myocardial tissues of the rats were collected,and the expressions of MMP-1 and TIMP-1 were analyzed by the gel imaging system with Western blotting method.Results:The expression of MMP-1 was significantly increased in the model group.The intervention of Guishen Tongmai Capsule of medium-dose and high-dose could significantly decrease the expression of MMP-1,which effect was more significant than that of Captopril.In terms of the expression of TIMP,it was the highest in the normal group,and the lowest in the model group;after the intervention of Captopril and Guishen Tongmai Capsule of medium-dose and high-dose,it was increased obviously,of which Guishen Tongmai Capusle of high-dose had the best result.Conclusion:Guishen Tongmai Capsule can decrease the expression of MMP-1 and increase the expression of TIMP-1,reduce the ratio of MMP-1/TIMP-1,delay the development of heart failure and inhibit or reverse the process of ventricular remodeling.
