摘 要:目的探讨提前给予脂多糖(LPS)对支气管哮喘(哮喘)预防的作用及相关免疫机制,为哮喘的预防和治疗提供理论依据。方法提前用LPS处理BALB/c背景6~8周小鼠,然后用卵清蛋白(OVA)联合铝佐剂诱导小鼠哮喘模型,阴性对照组用无菌生理盐水,分为以下4组:LPS给药1次组(LPS1组),LPS给药2次组(LPS2组),阴性对照组(NS组),OVA组。通过肺功能检测气道阻力、肺组织病理染色、支气管肺泡灌洗液分类细胞计数、酶联免疫吸附试验测Th2型细胞因子及白细胞介素6(IL-6)、IL-10,流式细胞术测肺组织调节性T细胞(Treg)比例等方法探索LPS对哮喘有无预防作用。结果LPS预处理小鼠在OVA诱导后,肺组织苏木精-伊红染色、过碘酸-希夫染色提示炎症减轻,炎细胞浸润减少,肺组织匀浆IL-5、IL-6、IL-13降低,LPS2组IL-10及Treg比例增高,与OVA组比较差异有统计学意义。结论LPS通过减少Th2型细胞因子及刺激Treg细胞增殖减轻OVA诱导的哮喘炎症反应。Objective:To explore the effect of early administration of lipopolysaccharide(LPS)on the prevention of bronchial asthma(asthma)and its related immune mechanism.Methods:BALB/c background,6-8 weeks old mice were intranasally administrated with LPS,then ovalbumin(OVA)and isopyknic aluminum were used to induce asthma model.The negative control group was treated with aseptic normal saline.The mice were divided into the following four groups:nasal drip once with LPS(LPS1 group),nasal drip twice with LPS(LPS2 group),negative control group(NS group),and positive control group(OVA group).We detected airway resistance,immunohistochemical staining of lung,and classified cell count of bronchoalveolar lavage fluid.We also tested Th2 cytokines,IL-6,and IL-10 by ELISA and Treg by flow cytometry to explore the preventive effect of LPS on asthma.Results:After OVA induction,hematoxylin-eosin staining and periodicacid-schiff staining in lung tissue of LPS pretreated mice showed that inflammation was alleviated,inflammatory cell infiltration was decreased.There was significant difference between OVA group and LPS1/LPS2 group.Level of interleukin-5(IL-5),IL-6 and IL-13 in lung homogenate were decreased in both LPS1 and LPS2 group compared with OVA group.Level of IL-10 and frequency of regulatory T cells(Treg)in LPS2 group were increased compared with OVA group.Conclusions:LPS attenuates the inflammation of asthma induced by OVA by down-regulation of Th2 cytokines and stimulating the frequency of Treg.
