摘 要:miRNA与畜禽的主要生产性能和生长发育密切相关,主要参与调控细胞分化增殖、组织器官生长发育等生理过程。miRNA表达具有组织特异性和发育的时序性,在细胞生长发育的不同阶段表达不同种类和数量的miRNA。本研究利用Stem-loop实时定量PCR检测miRNA-150和miRNA-143在阿勒泰羊体外胚胎5个发育阶段、瘤胃、肝脏等13种组织中的表达量,分析了miRNA-150和miRNA-143在阿勒泰羊不同组织中的表达特征。结果表明,绵羊miRNA-150和miRNA-143均为广泛性表达miRNA。miRNA-143在瘤胃、直肠、皱胃中显著高于其他组织的表达水平,miRNA-150在肝脏和骨骼肌中表达水平显著高于其他组织。胚胎时期miRNA-143在卵母细胞中显著高于其他时期的表达水平。miRNA-150在4细胞中表达水平显著高于其他时期。生物信息学分析显示,预测的miRNA-150的靶基因在mTOR信号通路、胰岛素信号通路和脂肪细胞因子信号通路富集;miRNA-143的靶基因在粘着连接、ECM受体相互作用通路和刺猬信号通路富集。总之,miRNA-150和miRNA-143为广泛性时序表达miRNA,可能参与绵羊诸多器官发育和胚胎发育相关的生物学过程的调控。miRNA is closely related to the main production performance and the growth and development,mainly involved in regulation of cell differentiation and proliferation and tissues and organs growth and development,.miRNA expression have scheduling of the development and tissue specificity,there are different kinds and quantities of miRNA expression.at various cell growth and development stages.This study used Stem-loop real-time quantitative PCR technique to detect miRNA-150 and miRNA-143 expression in Altay sheep’s embryos and thirteen kinds of tissues.The results showed that miRNA-150 and miRNA-143 are the generalized express in sheep.miRNA-143 expression level in the rumen,rectum,abomasum is significantly higher than other organizations,the miRNA-150 expression level in liver and skeletal muscle were significantly higher than other organizations.miRNA-143 expression level in the oocyte is significantly higher than other times.miRNA-150 expression level in four cell was significantly higher than other times.Bioinformatics analysis result showed that miRNA-150 target genes enrich in the mTOR signaling pathways,insulin signaling pathway and fat cytokine signaling pathway;miRNA-143 target genes enrich in adhesive connection pathway,ECM receptor interaction pathways and hedgehog signaling pathways.In short,miRNA-150 and miRNA-143 are the generalized and scheduled expression,may participate in biological control in many organ and embryo development.
