摘 要:目的:优化柴黄姜术合剂的提取工艺,为该制剂的生产标准化提供理论依据。方法:采用高效液相色谱-蒸发光散射检测器(high performance liquid chromatography-evaporative light scattering detector,HPLC-ELSD)法测定黄芪甲苷、柴胡皂苷a、柴胡皂苷b2含量,流动相乙腈(A)-水(B)梯度洗脱(0~17 min,30%A;17~18 min,30%~35%A;18~25 min,35%~40%A;25~26 min,30%~40%A),以黄芪甲苷、柴胡皂苷a、柴胡皂苷b2含量和干膏得率为评价指标,考察加水量、提取时间、提取次数对柴黄姜术合剂提取工艺的影响。结果:黄芪甲苷、柴胡皂苷a和柴胡皂苷b2含量的RSD分别为2.33%,2.14和1.87%,线性范围依次为1.20~12.03μg,0.67~6.78μg,0.88~8.82μg,平均加样回收率分别为98.87%,102.53%,99.37%。最佳提取工艺为8倍量水提取3次,每次1 h。结论:优化的提取工艺重复性好、稳定可靠、提取率高,适合柴黄姜术合剂的标准化生产。Objective:To optimize the extraction process of Chai Huang Jiang Zhu Mixture(柴黄姜术合剂,CHJZM)and provide theoretical basis for the production standardization of the preparation.Methods:High performance liquid chromatography evaporative light scattering detector(HPLC-ELSD)was used to determine the contents of astragaloside a,saikosaponin a and saikosaponin b2.The mobile phase was composed of acetonitrile(a)-water(b)and carried out according to the following gradient elution(0-17 min,30%A;17-18 min,30%-35%A;18-25 min,35%-40%A;25-26 min,30%-40%A).With astragaloside a,saikosaponin a,saikosaponin b2 contents and dry extract rate as evaluation indexes,the effects of water addition,extraction time and extraction frequency on the extraction process of CHJZM were investigated.Results:The RSD of astragaloside a,saikosaponin a and saikosaponin b2 were 2.33%,2.14 and 1.87% respectively.The linear range was 1.20-12.03μg,0.67-6.78μg,0.88-8.82μg respectively.The average recovery rate was 98.87%,102.53% and 99.37% respectively.The best extraction process is 8 times water with extraction for 3 times,1 hour each time.Conclusion:The optimized extraction process has good repeatability,stability,reliability and high extraction rate,which is suitable for the standardized production of CHJZM.
