摘 要:心房利钠肽(Atrial natriuretic peptide,ANP)是利钠肽家族成员之一,具有促进人脂肪细胞脂解的作用。研究旨在通过静脉滴注ANP观察其对绵羊血液中脂肪、碳水化合物和蛋白质代谢指标的影响。试验选取11月龄阿勒泰大尾羊10只,试验第1~4天为对照期,通过静脉滴注生理盐水100 mL/45 min;试验第5~8天为试验期,通过静脉按照25.0 ng/kg BW/min滴注合成的ANP生理盐水溶液100 mL/45min。试验的第2~4天和6~8天分别于滴注前(即0 min)、滴注后第15、30、45、60、90和120 min采集血液样本,用于测定血液生理生化指标。结果表明,静脉滴注ANP后,血液中甘油三酯(TG)的水平提高35.71%(P<0.001),游离脂肪酸(FFA)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)水平分别降低25.66%(P<0.001)、12.56%(P<0.001)、17.16%(P<0.001)和16.48%(P<0.001);血糖水平和乳酸脱氢酶(LDH)的活性分别降低7.83%(P=0.001)和10.37%(P<0.001);血液中谷丙转氨酶(GPT)和谷草转氨酶(GOT)活性分别降低10.46%(P=0.001)和14.02%(P<0.001),但血液尿素氮(BUN)水平无显著变化(P>0.05)。静脉滴注ANP可影响绵羊脂肪、蛋白质和糖代谢,具体机制需进一步研究。ANP(Atrial natriuretic peptide),one of the members of the family of natriuretic peptide,can promote human adipocytes lipolysis.The aim of this study was to investigate the effects of intravenous infusion of ANP on the metabolism indexes of lipid,carbohydrate and protein in blood of sheep.Ten Altay fat tailed sheep with 11 months of age were selected and self-controlled experimental design was used.In the control period(from d 1 to 4),100 mL saline was instilled through intravenous for 45 min.In the experimental period(from d 5 to 8),intravenous infusion of synthetic sheep ANP dissolved in 100 mL saline was instilled at rates of 25.0 ng/kg BW/min for 45 min.During d 2 to 4 and 6 to 8,the blood samples were collected before and after infusion of 15,30,45,60,90 and 120 min,and indexes of blood physiological and biochemical were determined.The results showed that the level of blood triglyceride(TG)increased by 35.71%(P<0.001)in ANP infusion period,and levels of free fatty acids(FFA),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)decreased by 25.66%(P<0.001),12.56%(P<0.001),17.16%(P<0.001)and 16.48%(P<0.001),respectively.Blood glucose and lactate dehydrogenase(LDH)decreased by 7.83%(P=0.001)and 10.37%(P<0.001).Activity of glutamic-pyruvic transaminase(GPT)and glutamic-oxalacetic transaminase(GOT)reduced by 10.46%(P=0.001)and 14.02%(P<0.001),but the level of blood urea nitrogen(BUN)showed no significant change(P>0.05).In summary,intravenous infusion of synthetic sheep ANP can affect the metabolism of lipid,protein and carbohydrate in sheep,but further researches are required on the specific mechanisms.
