摘 要:目的:通过研究阳和化岩汤对乳腺癌癌前病变细胞P53、Ki67的表达,探讨阳和化岩汤治疗乳腺癌癌前病变的机理。方法:将乳腺癌癌前病变MCF-10AT细胞分为阳和化岩汤组,三苯氧胺组,拆方温阳组、解毒组、活血组、化痰组及空白对照组,采用免疫细胞化学法和半定量RT-PCR法检测各组对Ki67、P53表达的影响。结果:治疗组与空白对照组相比,均能降低Ki67、突变型P53的表达,与空白对照组相比,阳和化岩汤组与三苯氧胺组(P<0.05)有统计学意义;与阳和化岩汤组相比,拆方组(P<0.05)有统计学意义。结论:阳和化岩汤可减缓肿瘤细胞增殖活性,达到延缓乳腺癌进一步发展的目的,其机制可能与降低Ki67、突变型P53的表达有关。Objective:To study the expressions of P53 and Ki67 in breast cancer precancerous cells intervened by Yanghe Huayan Decoction,and to explore the mechanism of Yanghe Huayan Decoction in treating breast cancer precancerous lesions.Methods:MCF-10 AT cells of breast cancer were divided into Yanghe Huayan Decoction group,Tamoxifen group,Wenyang group,Jiedu group,Huoxue group,Huatan group and the blank control group.The effects of each group on the expressions of Ki67 and P53 were detected by RT-PCR.Results:All the treatment groups could reduce the expressions of Ki67 and mutant P53.There were statistical differences between the blank control group and Yanghe Huayan Decoction group and between the blank control group and Tamoxifen group(P<0.05);there were statistical difference between Yanghe Huayan Decoction group and each sub-divided group(P<0.05).Conclusion:Yanghe Huayan Decoction can slow down the proliferation activity of tumor cells and delay the further development of breast cancer.The mechanism may be related to reducing the expressions of Ki67 and mutant P53.
