摘 要:目的:建立垂盆草的HPLC指纹图谱,对比不同产地垂盆草的差异,为垂盆草质量控制提供参考。方法:采用Welchrom C 18色谱柱(4.6 mm×250 mm,5μm),以0.1%乙酸水-乙腈为流动相进行梯度洗脱,流速为1.0 mL/min,柱温40℃,检测波长360 nm,建立垂盆草HPLC指纹图谱,采用相似度软件对指纹图谱进行分析,通过聚类分析和主成分分析,评价不同产地垂盆草的差异。结果:建立垂盆草特征指纹图谱,共确定7个共有峰,同产地样品相似度均在0.90以上,而不同产地之间相似度均小于0.9;聚类分析和主成分分析结果显示:四川产的4批垂盆草成分之间差异最小,而安徽产的3批垂盆草成分之间差异较大。结论:所建立的垂盆草HPLC特征指纹图谱方法简单,分离效果较好,可用于垂盆草的质量控制,为其相关研究提供指导;并且不同产地垂盆草化学成分差异较大。Objective:To establish an HPLC fingerprint method of Sedum sarmentosum Bunge,and contrast the difference of Sedum sarmentosum Bunge from different habitats,so as to provide a reference for the quality control.Methods:Welchrom C 18(4.6 mm×250 mm,5μm)column was adopted,the mobile phase consisted of 0.1%acetic acid water-acetonitrile with gradient elution at the flow rate of 1.0 mL·min-1,and the detection wavelength was 360 nm,the column temperature was 40℃.HPLC fingerprint of Sedum sarmentosum Bunge was established.Cluster analysis and principal component analysis were used to evaluate the chemical differences of Sedum sarmentosum Bunge from different habitats.Results:The fingerprint of Sedum sarmentosum Bunge was established,and included 7 common peaks.The similarity of samples from the same habitat was more than 0.90,while the similarity between different habitats was less than 0.90.Cluster analysis and principal component analysis showed that there was little difference among the four batches of S Sedum sarmentosum Bunge in Sichuan,but there was large difference among the three batches of Sedum sarmentosum Bunge in Anhui.Conclusion:The established HPLC fingerprint method is simple in good separation.It can be used for the quality control of Sedum sarmentosum Bunge and provide guidance for its related research.And the chemical constituents of Sedum sarmentosum Bunge from different habitats are different.
