四君子汤配方颗粒与传统汤剂对实验性大鼠脾虚改善作用的比较研究

摘　　要：目的:比较四君子汤配方颗粒与传统汤剂对实验性大鼠脾虚改善作用的差异。方法:90只SD大鼠随机分为9组:正常对照组、模型组、传统汤剂低(以生药材计1.485 g·kg^-1·d^-1)、中(以生药材计2.97 g·kg^-1·d^-1)、高(以生药材计5.94 g·kg^-1·d^-1)剂量组、配方颗粒汤剂低(以生药材计1.485 g·kg^-1·d^-1)、中(以生药材计2.97 g·kg^-1·d^-1)、高(以生药材计5.94 g·kg^-1·d^-1)剂量组、阳性对照组(枸橼酸莫沙必利分散片,5 mg·kg^-1·d^-1),每组10只。采用小承气汤法、劳倦法及饥饿法三因素复合法复制脾气虚证模型后,灌胃给药,15 d后处死大鼠。采用ELISA法测定血清中血清生长激素释放肽(Ghrelin)、胃泌素(GAS)和胃动素(MLT)的含量,甲苯胺蓝染色法对大鼠胃窦肥大细胞(MC)及MC脱颗粒计数。结果:与正常对照组比较,模型组大鼠血清中Ghrelin、GAS和MLT的含量显著降低(P<0.05),MC及MC脱颗粒数量显著升高(P<0.05);与模型组比较,四君子汤配方颗粒与传统汤剂各组血清中Ghrelin、GAS和MLT的含量显著升高(P<0.05),MC及MC脱颗粒数量显著降低(P<0.05),配方颗粒与传统汤剂两者之间比较差异无统计学意义(P>0.05)。结论:四君子汤配方颗粒与传统汤剂均能改善对实验性大鼠脾虚症状,两者作用无显著差异。Objective:To compare the improvement of spleen deficiency between the formula granules and the traditional decoction of Sijunzi decoction in experimental rats.Methods:Totally 90 SD rats were randomly divided into nine groups:the normal control group,the model group,the traditional decoction low(based on raw medicinal materials,1.485 g·kg^-1·d^-1),medium(based on raw medicinal materials,2.97 g·kg^-1·d^-1)and high(based on raw medicinal materials,5.94 g·kg^-1·d^-1)dose groups,the formula granules low(based on raw medicinal materials,1.485 g·kg^-1·d^-1),medium(based on raw medicinal materials,2.97 g·kg^-1·d^-1)and high(based on raw medicinal materials,5.94 g·kg^-1·d^-1)dose groups,and the positive control group(moxapride citrate dispersible tablets,5 mg·kg^-1·d^-1)with 10 rats in each group.The model of spleen deficiency syndrome was established by using three factors of Xiaochengqi decoction method,overstrain method and starvation.After the molding,the drug groups were intragastrically administered with corresponding drug once per day consecutively for 15 days.After the experiment,the contents of serum Ghrelin,GAS and MLT were measured by ELISA,and the MC and MC degranulation stained with toluidine blue were also observed.Results:The contents of serum Ghrelin,GAS and MLT in the model group were significantly decreased compared with those in the normal group(P<0.05),while the number of MC and MC degranulation increased significantly(P<0.05).Meanwhile,compared with those in the model group,the contents of serum Ghrelin,GAS and MLT were significantly increased both in the groups of the formula granules and the traditional decoction of Sijunzi decoction(P<0.05),while the number of MC and MC degranulation were decreased significantly(P<0.05).There were no significant differences in above indics between the formula granules and the traditional decoction(P>0.05).Conclusion:Both the formula granules and the traditional decoction of Sijunzi decoction can improve spleen deficiency symptoms of experimental rats
