菜芙蓉多糖的表征与生物活性

摘　　要：以菜芙蓉花醇提后的残留物为原料,通过水提醇沉法提取菜芙蓉多糖,测定多糖总糖含量及得率。经DEAE-52柱层析分离纯化菜芙蓉多糖〔Abelmoschus manihot(L.)Medic polysaccharide,AMP〕,确定AMP单糖组成,考察AMP的体外抗氧化能力,并对AMP保护质粒DNA及抑制肝癌细胞Hep G2的增殖作用进行了考察。结果表明,菜芙蓉花多糖得率为13.47%,总糖含量为97.43%,AMP主要由鼠李糖、葡萄糖、阿拉伯糖、岩藻糖组成,其物质的量比为1.44∶5.44∶1.00∶5.37,体外清除自由基能力表现顺序为·O2^->ABTS^+·>DPPH·>·OH>NO·,8 g/L AMP抑制H2O2诱导的DNA氧化损伤效果最好,在0.1250~5.000 g/L范围内,随着AMP质量浓度的增加Hep G2细胞的增殖抑制率逐渐增大,说明AMP具有抗肿瘤活性。Polysaccharide was extracted from the ethanol-extracted debris of Abelmoschus manihot(L.)Medic flowers by water extraction and alcohol precipitation method.The total sugar content and extraction rate of polysaccharide were determined.Abelmoschus manihot(L.)Medic polysaccharide(AMP)was separated and purified by DEAE-52 column chromatography and the composition of monosaccharide of AMP was determined.The bioactivities of AMP,including antioxidant activity in vitro,DNA protecting activity and hepatic cancer cell HepG2 growth inhibitory effect were investigated.The results showed that the extraction rate reached 13.47%and the content of total sugar was 97.43%.AMP was mainly composed of rhamnose,glucose,arabinose and fucose at a molar ratio of 1.44∶5.44∶1.00∶5.37.The order of radicals scavenging capacities of AMP was·O2^->ABTS^+·>DPPH·>·OH>NO·,suggesting AMP had certain antioxidant activity in vitro.AMP(8 g/L)had the best inhibition effect on inhibiting DNA oxidative damage induced by H2O2.In the range of 0.1250 to 5.000 g/L,with the increase of AMP mass concentration,the inhibitory rate of HepG2 cells proliferation gradually increased,indicating that AMP had potential antitumor activity.
