摘 要:目的探索血竭素高氯酸盐(dracorhodin perchlorate,DP)对体外培养的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)的增殖、迁移和管腔样结构形成的影响。方法自2019年1—6月,取体外培养的第4~6代HUVECs,分为对照组和观察组。对照组在ECM培养基中添加DMSO;观察组添加不同浓度的(0.1、1.0、5.0、10.0和20.0 mmol/L)的DP。处理8 h后,采用小管形成实验检测DP对HUVECs管腔样结构形成的影响;48 h后,采用MTS试剂盒和划痕实验分别检测DP对HUVECs的增殖和迁移的影响。结果与对照组相比,采用5.0、10.0和20.0 mmol/L的DP进行处理,可显著抑制HUVECs的增殖和迁移(P<0.01),同时可显著抑制管腔样结构的形成(P<0.05)。结论DP对HUVECs的增殖和迁移以及管腔样结构形成能力,具有显著的抑制作用。Objective To explore the effect of dracorhodin perchlorate(DP)on proliferation,migration and tube-like structure formation of human umbilical vein endothelial cells(HUVECs)in vitro.Methods From January to June 2019,the HUVECs cultured in vitro from passage 4 to 6 were divided into the control group and the observation group.The DMSO was added to the ECMmedium in the control group and DP of different concentrations(0.1,1.0,5.0,10.0 and 20.0 mmol/L)was added in the observation group.After treatment of 8 hours,the effect of DP on tube-like structure formation of HUVECs was explored by tube formation assay.After treatment of 48 hours,the effect ofDP on proliferation and migration ofHUVECs was detected byMTS kit and scratch assay,respectively.Results Compared with the control group,the proliferation and migration of HUVECs could be significantly inhibited after treatment by DP of 5.0,10.0 and 20.0 mmol/L(P<0.01).At the same time,the formation of tube-like structure was significantly inhibited(P<0.05).Conclusion The proliferation,migration and formation of tube-like structure ofHUVECs could be significantly inhibited by DP.
