续断对兔膝骨关节炎模型滑膜细胞凋亡蛋白Bcl-2和Fas-L表达影响的实验研究

摘　　要：目的探讨中药续断对兔膝骨关节炎(knee osteoarthritis,KOA)模型滑膜细胞凋亡蛋白Bcl-2和Fas-L影响。方法采用随机数字法将动物分为3组,正常组、对照组和续断组各12只。对3组模型滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率进行比较。结果治疗前滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率续断组与对照组差异无统计学意义(P> 0. 05),续断组均显著高于正常组(t值分别为10. 61、10. 51,P <0. 01);对照组均显著高于正常组(t值分别为9. 43、7. 73,P <0. 01);治疗4周后滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率比较,续断组显著低于对照组(t值分别为9. 23、7. 35,P <0. 01),而与正常组差异无统计学意义(P> 0. 05);对照组均显著高于正常组(t值分别为10. 27、7. 86,P <0. 01);滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率与续断治疗呈显著负相关(r值分别为-0. 83、-0. 94,P <0. 01)。滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率与续断治疗时间呈显著正相关(r值分别为0. 86、0. 89,P <0. 01)。结论续断可降低KOA滑膜细胞凋亡蛋白Bcl-2和Fas-L表达阳性细胞率。Objective To observe the effect of radix dipsaci on synovial apoptotic protein Bcl-2 and Fas-L expression in rabbit models of knee osteoarthritis. Methods The research objects were divided into 3 groups,including normal group,control group and radix dipsaci group by stratified randomization,with 12 rabbits in each group. The positive cell rates of Bcl-2 and Fas-L in synovium were detected in the three groups. Results There was no statistical significance on the difference in the positive cell rates of Bcl-2 and Fas-L between the intermittent group and the control group before treatment( P > 0. 05),and the intermittent group was significantly higher than the normal group( t = 10. 61,t = 10. 51,P < 0. 01);the control group was significantly higher than the normal group( t = 9. 43,t = 7. 73,P < 0. 01);after 4 weeks of treatment,the positive cell rates of Bcl-2 and Fas-L in the discontinuous group were significantly lower than that in the control group( t = 9. 23,t = 7. 35,P <0. 01),but not statistically significant compared to the normal group( P > 0. 05);the positive cell rates of Bcl-2 and Fas-L in the control group were significantly higher than that in the normal group( t = 10. 27,t = 7. 86,P < 0. 01). The positive cell rates of Bcl-2 and Fas-L were negatively correlated with intermittent treatment( r =-0. 83,r =-0. 94,P < 0. 01). The positive cell rates of Bcl-2 and Fas-L were positively correlated with the duration of intermittent treatment( r = 0. 86,r = 0. 89,P < 0. 01). Conclusion Radix dipsaci can reduce the positive cell rates of Bcl-2 and Fas-L in KOA synovial cells.
