摘  要:CCCH型锌指蛋白是植物中广泛存在的一类转录因子,在植物生长发育、胁迫应答等多种生物过程中发挥重要的调控作用。本研究从刚毛柽柳(Tamarix hispida)中克隆获得一条CCCH型锌指蛋白基因,命名为ThZFP1。该基因开放阅读框(ORF)长870 bp,编码290个氨基酸,推测蛋白质分子质量为30.38 kDa,理论等电点为5.10。ThZFP1属于疏水性蛋白。进化树分析结果表明, ThZFP1与茶树ZFP的亲缘关系最近。实时荧光定量PCR分析结果显示,在NaCl、PEG和ABA处理下,ThZFP1基因的表达量明显上调,表明ThZFP1基因能对高盐、干旱胁迫和ABA处理做出应答。CCCH-type zinc finger protein is a class of transcription factor in plant, which plays an important regulatory role in various biological processes such as grow, development and stress response. In this study, a zinc finger protein gene was cloned from Tamarix hispida cDNA library, named ThZFP1. The opening reading frame(ORF) length of ThZFP1 was 870 bp, encoding 290 amino acid. The relative molecular weight of ThZFP1 protein was 30.38 kDa, with isoelectric point 5.10. ThZFP1 is a hydrophobic protein and may be localized in the nucleus. Quantitative real-time PCR showed that the expression level of ThZFP1 gene was significantly up-regulated under NaCl, PEG and ABA treatments, indicating that ThZFP1 gene responded to salt and drought stress and ABA hormone treatment.
