摘 要:目的研究黄芪对人B细胞淋巴瘤Raji细胞增殖、凋亡的影响及潜在机制。方法用200、400、600和800μg/ml黄芪处理人B细胞淋巴瘤Raji细胞24、48、72 h,MTT法测定不同浓度黄芪对Raji细胞的增殖活性的影响;流式细胞术和实时荧光定量PCR(qRT-PCR)分别测定600μg/ml黄芪处理后Raji细胞凋亡率和细胞中肿瘤坏死因子α诱导蛋白3(TNFAIP3)mRNA的表达;免疫印迹检测Raji细胞中TNFAIP3、Cyclin D1、P21、Bax和Bcl-2蛋白表达。结果黄芪处理后,Raji细胞中Cyclin D1、Bcl-2表达下调,p21、Bax表达上调,可明显抑制细胞增殖,诱导细胞凋亡,且具有剂量依赖性(P<0.05);黄芪可促进Raji细胞中的TNFAIP3的表达,抑制TNFAIP3可逆转黄芪对Raji细胞增殖和凋亡的作用。结论黄芪通过促进TNFAIP3表达抑制人B细胞淋巴瘤Raji细胞增殖并促进细胞凋亡。黄芪对人B细胞淋巴瘤具有潜在治疗作用。Objective To investigate the effect of astragalus on proliferation and apoptosis of human B cell lymphoma cells and its potential mechanism.Methods Human B cell lymphoma Raji cells were treated for 24,48 and 72 h with the 200,400,600 and 800μg/ml of astragalus.The effect of astragalus at different concentrations on the proliferation of Raji cells was determined by MTT assay.The cell apoptosis rate and tumor necrosis factor alpha-induced protein 3(TNFAIP3)mRNA expression in Raji cells after treatment with 600μg/ml astragalus were determined by flow cytometry and qRT-PCR.The protein expression levels of TNFAIP3,Cyclin D1,P21,Bax and Bcl-2 in Raji cells were detected by Western blotting.Results Treatment with astragalus resulted in down-regulated expression of Cyclin D1 and Bcl-2,up-regulated expression of p21 and Bax,inhibited proliferation and promoted apoptosis of Raji cells in a dose-dependent manner(P<0.05).Astragalus promoted the expression of TNFAIP3 in Raji cells.Inhibition of TNFAIP3 reversed the effect of astragalus on the proliferation and apoptosis of Raji cells.Conclusion Astragalus inhibits the proliferation and promotes the apoptosis of human B cell lymphoma Raji cells by enhancing the expression of TNFAIP3,and therefore offers therapeutic potential for human B cell lymphoma.
