摘  要:目的探索羟基积雪草酸(MA)对人舌癌Tca8113细胞生长、凋亡的作用及机制。方法体外培养人舌癌Tca8113细胞,并将细胞分为对照组和低、中、高剂量实验组。对照组不加MA;低、中、高剂量实验组分别给予40,80和120μmol·L^-1 MA。用噻唑蓝法检测Tca8113细胞24 h的细胞生存率,用Annexin V-FITC/PI双染法检测Tca8113细胞12 h的凋亡率,用Western Blotting法检测Bcl-2、p53、Bax和Cleaved Caspase-3的表达。结果低、高剂量实验组和对照组的24 h细胞生存率分别为(89.13±4.54)%,(57.86±6.09)%和(100.00±0)%,12 h凋亡率分别为(5.31±0.12)%,(20.86±0.45)%和(1.94±0.87)%,Bax蛋白分别为(0.20±0.06),(0.43±0.08)和(0.08±0.04),p53蛋白分别为(0.29±0.08),(0.49±0.06)和(0.14±0.08),Cleaved Caspase-3蛋白分别为(0.24±0.04),(0.52±0.02)和(0.16±0.04),Bcl-2蛋白分别为(0.71±0.12),(0.37±0.14)和(0.80±0.08),高剂量实验组的上述指标与对照组比较,差异均有统计学意义(均P<0.05)。结论 MA能抑制人舌癌Tca8113细胞增殖并促其凋亡,其机制可能与上调Bax、p53和Cleaved Caspase-3的表达及下调Bcl-2的表达有关。Objective To investigate the effect and mechanism of Madecasis acid(MA)on the growth and apoptosis of human tongue cancer Tca8113 cells.Methods Human tongue cancer Tca8113 cells were cultured in vitro,and then divided into control group and experimental-L,-M,-H groups.Control group did not add MA.Experimental-L,-M,-H groups were given 40,80,and 120μmol·L^-1 MA,respectively.The cell viability rate of Tca8113 cells in 24 hours was measured by methyl thiazolyl tetrazolium.The apoptosis rate of Tca8113 cell in 12 hours was measured by Annexin V-FITC/PI double staining method.The protein expressions of Bcl-2,p53,Bax and Cleaved Caspase-3 were detected by Western Blotting.Results The cell viability rates of control group and experimental-L,-H groups in 24 hours were(89.13±4.54)%,(57.86±6.09)%and(100.00±0)%,apoptosis rates in 12 hours of the above 3 groups were(5.31±0.12)%,(20.86±0.45)%and(1.94±0.87)%,Bax protein of the above 3 groups were(0.20±0.06),(0.43±0.08)and(0.08±0.35),p53 protein of the above 3 groups were(0.29±0.08),(0.49±0.06)and(0.14±0.08),Cleaved Caspase-3 protein of the above 3 groups were(0.24±0.04),(0.52±0.02)and(0.16±0.04),Bcl-2 protein of the above 3 groups were(0.71±0.12),(0.37±0.14)and(0.80±0.08).There were significant differences in the above-mentioned indexes between experimental-H group and control group(all P<0.05).Conclusion MA can inhibit the proliferation and promote apoptosis of human tongue cancer Tca8113 cells,which may be related to up-regulating the expression of Bax,p53 and Cleaved Caspase-3 and down-regulating the expression of Bcl-2.
