摘  要:目的:研究小鼠中脑不同区域多巴胺能神经元的电生理特性,为探究其在运动性疾病及焦虑抑郁中的机制提供研究基础.方法:8只成年DAT-tdtomato小鼠,麻醉后急性断头取脑,冠状切片(350μm),应用脑片膜片钳全细胞记录技术记录中脑黑质致密部(SNc)、腹侧被盖区(VTA)内侧和外侧多巴胺能神经元被动膜特性及主动膜特性,统计分析不同区域多巴胺能神经元电生理特性的差异.结果:SNc区和VTA外侧区多巴胺能神经元的膜电容(Cm)和后超极化(AHP)的幅度显著高于VTA内侧区(P<0.001).SNc区和VTA外侧区多巴胺能神经元Sag的幅度显著高于VTA内侧区(P<0.001),但是VTA内侧区多巴胺能神经元动作电位的发放数目(spike number)显著高于SNc区和VTA外侧区(P<0.001).结论:SNc区和VTA外侧区多巴胺能神经元的电生理特性与VTA内侧区明显不同,但SNc区和VTA外侧区多巴胺能神经元电生理特性也存在差异.Objective:To study the electrophysiological characteristics of dopaminergic neurons in different regions of the midbrain in mice,and to provide a basic study on the properties of dopaminergic neurons that may be valuable for their different roles in motor diseases,anxiety or depression.Methods:Eight DAT-tdtomato transgenic mice were used to prepare coronal slices(350μm)after anesthesia.Whole-cell patch recording was employed to record the passive membrane properties and active membrane properties of dopaminergic neurons in substantia nigra compact(SNc)and the medial and lateral ventral tegmental area(VTA).The differences of electrophysiological characteristics in dopamin­ergic neurons were analyzed in different region.Results:The magnitude of membrane capacitance(Cm)and after hyperpolarization potential(AHP)of dopaminergic neurons in SNc and lateral VTA regions were.significantly higher than that in medial VTA(P<0.001).Meanwhile,the Sag amplitude of dopaminergic neurons in SNc and lateral VTA were significantly higher than that in medial VTA(P<0.001).However,the spike number of dopaminergic neurons in medial VTA was significantly higher than those in SNc and lateral VTA region(P<0.001).Conclusion:The electrophysiological characteristics of dopaminergic neurons in the SNc and lateral VTA were significantly different from those in the medial VTA,but there were also differences between the SNc and lateral VTA.
