摘 要:目的观察可降解生物医学材料左旋聚乳酸(poly-L-lactic acid,PLLA)对巨噬细胞极化的体外影响,并探讨姜黄素改善PLLA植入后生物相容性的作用.方法采用PLLA(PLLA组)、姜黄素(姜黄素组)及其两者共同作用(PLLA+姜黄素组)于人类THP-1单核细胞分化而来的巨噬细胞,通过CCK8试剂盒检测PLLA和姜黄素对巨噬细胞的细胞毒性,通过ELISA和RT-qPCR检测各组巨噬细胞M1和M2的细胞因子[白细胞介素(IL)-6、肿瘤坏死因子-α(TNF-α)和IL-10]的表达水平和基因转录水平检测.结果与对照组比较,PLLA组巨噬细胞炎症型M1细胞因子TNF-α、IL-6表达和转录水平显著升高,修复抗炎型M2细胞因子IL-10的表达水平升高但转录水平下降;与PLLA组比较,PLLA+姜黄素组的M1型细胞因子TNF-α、IL-6表达和转录水平下降,M2型细胞因子IL-10表达和转录水平上升.结论PLLA促使体外培养的巨噬细胞向炎症型M1转化为主,且姜黄素促使PLLA作用下的巨噬细胞向M2型转化.Objective To investigate the effect of biodegradable biomaterials, as poly-L-lactic acid(PLLA) on macrophage polarization in vitro, and to investigate the effect of curcumin on the biocompatibility of PLLA after implantation. Methods PLLA, curcumin and their mixture were used to differentiate macrophages from human THP-1 monocytes in vitro. The expression level and gene transcription of cytokines(IL-6, TNF-α and IL-10) of macrophages M1 and M2 were detected by ELISA and RT-qPCR. Results Compared with the control group, PLLA significantly increased expression lever and gene transcription lever of macrophages M1 cytokines IL-6 and TNF-α, and increased the expression lever but decreased the gene transcription lever of IL-10 from macrophage M2. When curcumin was added, M1 type cytokines decreased and M2 cytokines increased in both expression lever and transcription lever.Conclusion In vitro, PLLA can promote macrophages to transform into inflammatory M1 mainly, then developed macrophages to transform from inflammatory M1 to repair anti-inflammatory M2 after curcumin′s addition.
